土党参多糖分离纯化及抗氧化能力分析 被引量:4

Isolation, purification and antioxidant activity of polysaccharide from Codonopsis Lanceolata

摘要 从土党参中分离纯化多糖并进行体外抗氧化能力分析。试验通过微波辅助提取、乙醇沉淀、Sevag去蛋白质、透析及DEAE-Cellulose 52柱层析、Sephadex G-100柱层析分离纯化土党参多糖,采用比色法测定其含量。以Vc为对照,检测多糖组分对DPPH·、·OH和O_2^-·自由基的清除能力,得出土党参多糖单组分的体外抗氧化能力。试验结果:微波提取土党参多糖得率为12.58%,在4次脱蛋白后,蛋白脱出率达到55.2%,多糖的损失率为26.4%。经柱层析分离纯化得到2个含量较高的多糖组分CLP-1和CLP-2。它们均对DPPH·、·OH和O_2^-·自由基有一定抑制作用并表现出量效关系。 The polysaccharide from Codonopsis Lanceolata was isolated and purified and analysis of antioxidant activity in vitro. Test was that assisted extraction by microwave, ethanol precipitation, removed protein by sevage, dialysis and DEAE-Cellulose 52 column chromatography, Sephadex G-100 column chromatography to isolate and purify polysaccharides in Codonopsis Lanceolata. Colorimetric method was used to determine its content. Taking vitamin C as positive control group, detection of removal ability in polysaccharide component to DPPH·, ·OH, O2^-· free radical to reach that external oxidation capability by a single group of polysaccharide in Codonopsis Lanceolata. Result was that rate of 12.58% by microwave to extract polysaccharides in Codonopsis Lanceolata. After 4 times separation of protein, extrusion rate was 55.2% of protein, loss rate was 26.4% of polysaccharide. Two contents of polysaccharide CLP-1 and CLP-2 were isolated and purified by column chromatography. They both had some inhibitory action and showed dose-effect relationship on DPPH·, ·OH, O2^-· free radical.
