杨树中有机酸的高效液相色谱测定

摘要 有机酸是动植物代谢过程中重要的中间产物,其含量多寡和变化可反映动植物体应激或胁迫情况下的抗性行为或自我调剂能力,为了进一步了解杨树在逆境中的一系列应激反应与有机酸变化的关系,本文建立了高效液相色谱法测定杨树中的草酸、苹果酸、酒石酸、柠檬酸和琥珀酸五种有机酸的方法。样品经前处理后,用反相色谱柱,以DH3.0的3%CH3OH+97%H20为流动相,流速0.8mL/min,检测波长210nm下进行分离、测定。本方法五种有机酸回收率在94.50%-114.05%之间,定量限在0.007-0.73mg/L之间,相对标准偏差为0.5%-0.7%。 Organic acid is important intermediates in the metabolic processes of plants and annimals. The content of the amount and the change can reflect the dynamic plant stress or stress resistance behavior of self-adjustment ability. In order to further understand the changes of poplar in the face of adversity, a series of stress reaction and organic acid system, the method for the determination of oxalic acid, malic acid, tartaric acid, citric acid and succinic acids in poplar was established by using high performance liquid chromatography. After pre-treating, the samples were determined on a reverce with 3% CH3OH + 97% HzO as mobile phase, flow rate was 0.8ml/min, detection wavelength was 210nm. The recoveries were 94.30% -114.05% and the relative standard deviation was 0.3% -0.7%, the limit of quantitation was 0.007- 0.73mg / L
