摘要
目的 在体外培养的人鼻腔上皮细胞模型上,探讨两性霉素B对鼻黏膜上皮细胞纤毛摆动频率(ciliary beat frequency,CBF)的影响.方法 体外培养6例人钩突和/或下鼻甲组织的黏膜上皮细胞,培养至7-10 d时给予不同质量浓度(0.25、0.5、1.0、2.0、4.0μg/ml)的两性霉素B处理,以生理盐水(normal saline,NS)作为对照组.分别测量加药前以及加药处理后20 min内的CBF.结果 NS对照组CBF在20 min内无明显变化(F=0.351,P>0.05).0.25、0.5、1.0 μg/ml两性霉素B处理组CBF在加药20 min内较加药前CBF无明显变化(F值分别为0.286、0.468、0.383,P值均>0.05).2.0μg/ml两性霉素B处理组CBF随时间延长逐渐降低,(F=1.908,P<0.05),其中加药7 min后,CBF降低效果显著;4.0 μg/ml两性霉素B处理组CBF随作用时间延长逐渐降低,(F=8.223,P<0.05),其中加药6 min后,CBF降低效果显著.结论 两性霉素B鼻腔局部应用可引起浓度依赖性CBF降低,在鼻腔局部应用时应考虑其对纤毛功能的影响.
Objective To investigate the effects of Amphotericin B on ciliary beat frequency (CBF) in in vitro cultured human nasal epithelial cells. Methods Human nasal epithelial cells derived from uncinate process or inferior turbinate of six patients were cultured in vitro, and treated with different concentrations of Amphotericin B (0. 25,0. 5,1.0,2. 0,4. 0 μg/ml)over a peroid of 7 - 10 d; normal saline (NS) was used as a negative control. CBF was detected before treatment and recorded continuously for 20 rain after treatment. Results CBF of epithelial cells in normal saline ( NS ) control group showed no significant changes in 20 rain ( F = 0. 351, P 〉 0. 05 ) ; Amphotericin B at 0. 25 μg/ml ( F = 0. 286, P 〉 0. 05 ), 0.5 μg/ml( F = 0. 468, P 〉 0. 05 ) or 1.0 μg/ml ( F = 0. 383, P 〉 0. 05 ) did not affect epithelial CBF in 20 rain measurement period; Amphoteriein B at 2. 0 μg/ml decreased epithelial CBF with time( F = 1. 908,P 〈 0. 05), significant changes were observed after 7 rain of drug treatment; Amphotericin B at 4. 0 μg/ml decreased epithelial CBF with time ( F = 8. 223, P 〈 0. 05 ) , significant changes were observed after 6 rain of drug treatment. Conclusions Amphoteriein B caused a dose-dependent decrease of human nasal CBF. Therefore, the influence of this drug on ciliary function should be considered when nasal topically applied.
