山慈菇提取物对结肠癌HT29细胞凋亡的影响 被引量:17

摘要 目的:研究山慈菇提取物对人结肠癌HT29细胞增殖及凋亡的影响。方法:采用MTT法检测山慈菇提取物对人结肠癌HT29细胞抑制率;流式细胞仪检测细胞凋亡率;Western blot法检测细胞凋亡相关蛋白Cyt-C、Bcl-2、Bax、Caspase-3的影响。结果:不同浓度的山慈菇提取物对HT29细胞增殖均有抑制作用,且抑制率呈时间、剂量依赖关系;山慈菇提取物干预后诱导HT29细胞凋亡(P<0.01)差异有统计学意义;山慈菇提取物诱导细胞凋亡过程中的CytC、Bax、Caspase-3蛋白表达明显增强,Bcl-2蛋白表达明显减弱(P<0.05)。结论:山慈菇提取物对人结肠癌HT29细胞有明显的促凋亡作用,其机制可能与上调Cyt-C、Bax、Caspase-3下调Bcl-2蛋白的表达有关。 Objective To investigate the effects of Cremastra appendiculata extract on proliferation and apoptosis in human colon cancer HT29 cells. Methods Using the MTF assay to determine the cell inhibition rates on human colon cancer HT29 cells; the flow cytometry to determine the apoptosis rates; Western blot technology to determine the expressions of Cyt - C, Bcl - 2, Bax and Caspase -3 protein. Results Cremastra appendieulata extract inhibited the human colon cancer HT29 cells in a time dependent and dose dependent manner, the apoptosis rates of the cells were increased after being cultured by Cremastra appendiculata extract; the expressions of Cyt - C, Bax and Caspase - 3 protein were increased and the expression of Bcl - 2 protein was decreased ( P 〈 0. 05 ). Condusions Cremastra appendiculata extract can induce the apoptosis of HT29 cells by a mechanism that may be related to increase the expression of Cyt - C, Bax and Caspase - 3 protein as well as decrease the expression of Bcl - 2 protein.
