摘要
为了探讨将白细胞分化抗原14(Clusterofdifferentiationantigen14,CD14)作为繁殖性能遗传标记的可行性,试验采用PCR-RPLP方法对64头梅山猪种猪CD14基因-61位点的多态性进行检测,同时利用广义线性模型(GLM)分析其与繁殖性能间的关系。结果表明:梅山猪CD14基因-61位点经BamHI酶切后可检测到AA、AG、GG3种基因型,其频率分别为0.406,0.500,0.094;遗传多态性分析表明,梅山猪CD14基因的多态信息含量(PIC)为0.349,介于0.25~0.5之间,表现为中度多态;关联分析结果表明,在初产母猪中,GG基因型个体的总产仔数显著低于AA基因型和AG基因型(P〈0.05),且GG基因型个体的产活仔数、断奶仔猪数、初生重和断奶重均低于AA基因型和AG基因型,但差异均不显著(P〉0.05);在经产母猪中,各性状之间差异不显著(P〉0,05).CD14基因-61位点的多态性对梅山猪初产母猪的繁殖性能有显著的遗传效应,但其对经产母猪的繁殖性能没有显著影响。说明梅山猪CD14基因-61位点并不能作为繁殖性能的分子遗传标记,但如果基于梅山猪CD14基因-61位点对于梅山猪的一般抗病力进行分子选育,不会对梅山猪的繁殖性能产生显著的不利影响。
To investigate the feasibility of leukocyte differentiation antigen 14- cluster of differentiation antigen 14( CD14) as a genetic marker of reproductive performance,the polymorphisms at locus- 61 of CD14 genes in 64 Meishan studpigs were detected using PCR- RFLP method.Meanwhile,the relationship between different genotypes and the reproductive performance was analyzed with generalized linear models( GLM).The results showed that three kinds of genotypes including AA,AG and GG were detected after the- 61 locus of CD14 gene of Meishan pigs was digested by Bam H Ⅰ,whose gene frequency was 0. 406,0. 500,and 0. 094,respectively. Genetic polymorphism analysis showed that the polymorphic information content( PIC) value of CD14 gene in Meishan pigs was 0. 349 and between 0. 25 to 0. 5,which displayed moderate polymorphism. The results of correlation analysis showed that the total number born of individuals with genotype GG was significantly lower( P < 0. 05) than that of individuals with genotypes AA and AG in primiparous sows. At the same time,the number born alive,weaningpiglets,birth weight and weaning weight of individuals with genotype GG were all lower than those of individuals with genotypes AA and AG in primiparous sows,but there were no significant differences( P > 0. 05) between them. However,there were no significant differences( P > 0. 05) among all traits in multiparous sows. The polymorphism at- 61 locus of CD14 gene had a significant genetic effect on the productive performance of primiparous Meishan sows,but it had a significant effect on the productive performance of multiparous Meishan pigs. The results indicate that the locus- 61 of CD14 genes in Meishan pigs can't serve as the molecular genetic marker of reproductive performance,but it will not produce a significant adverse impact if molecular breeding of the general disease resistance of Meishan pigs is conducted based on the- 61 locus of CD14 gene in Meishan pigs.
