益气温阳通瘀化痰法对心力衰竭大鼠心肌能量代谢作用机制的研究 被引量:8

摘要 目的探讨益气温阳通瘀化痰法代表方芪红散对心力衰竭大鼠心肌能量代谢及心功能的影响,揭示芪红散治疗心力衰竭作用机制。方法通过建立大鼠心衰模型,采用芪红散进行干预实验,将心衰大鼠随机分为模型组、芪红散低剂量组(QHD)、芪红散中剂量组(QHZ)、芪红散高剂量组(QHG)和曲美他嗪组,灌胃给药12周后,测定各组大鼠Na^+-K^+-ATPase、Ca^(2+)-ATPase指标,并测量各组大鼠左室收缩末内径(LVESD)、左室舒张末内径(LVEDD)、左室短轴缩短率(FS)、左室射血分数(LVEF)。结果治疗12周后,模型组、QHD组、QHZ组、QHG组及曲美他嗪组与空白组比较,Na^+-K^+-ATPase、Ca^(2+)-ATPase均明显升高(P<0.05);与模型组比较,QHD组、QHZ组、QHG组及曲美他嗪组Na^+-K^+-ATPase、Ca^(2+)-ATPase均显著升高(P<0.05)。模型组、QHD组、QHZ组、QHG组及曲美他嗪组与空白组相比,LVESD增大,LVEF降低,差异有统计学意义(P<0.05);QHZ组、QHG组及曲美他嗪组与模型组相比,LVEDD、LVESD降低,LVEF增加,差异有统计学意义(P<0.05)。结论芪红散可以通过提高心力衰竭大鼠Na^+-K^+-ATPase、Ca^(2+)-ATPase酶含量,降低LVEDD、LVESD,增加LVEF,从而保护心肌,改善心力衰竭大鼠症状,提高心功能。 Objective To explore the mechanism of nourishing qi, warming yang,activating blood and clearing phlegm therapies onthe myocardial energy metabolism in rats with heart failure. Methods The rat model with heart failure was established. The rats withheart failure were randomly divided into model group, Qihong granule ( QHG)low dose group, QHG medium dose group, QHG highdose group and the trimetazidine group, determination of irrigation to 12 weeks after heart failure.Na^+ K^+ ATPase, Ca^2+ ATPasewere tested. Left ventricular end systolic dimension ( LVESD) , left ventricular end diastolic diameter ( LVEDD) , left ventricular shortaxis shortening rate ( FS) , left ventricular ejection fraction ( LVEF) were measured. Results After 12 weeks of treatment,the levelsof Na^+ K^+ ATPase, Ca^2+ ATPase were significantly increased in model group, QHG group and the trimetazidine group comparedwith blank group( P 〈0.05) . Compared with model group,the levels of Na^+ K^+ ATPase, Ca^2+ ATPase were significantly increased in QHG group and the trimetazidine group ( P 〈0.05) . Compared with blank group, LVESD increased, LVEF decreased inmodel group, QHG group and the trimetazidine group( P 〈0.05) . Compared with model group,LVEDD, LVESD decreased, LVEF increased in QHG medium dose group, QHG high dose group and the trimetazidine group( P 〈0.05) . Conclusion QHG can improvethe content of Na^+ K^+ ATPase, Ca^2+ ATPase, reduce the LVEDD, LVESD, increase the LVEF, thus protecting cardiac muscle, improve the symptoms of heart failure in rats, improve heart function in rats with heart failure.
