摘要
目的 探讨紧密连接蛋白Claudin-4在子宫内膜异位症(内异症)在位、异位内膜与正常内膜中的表达,及其在内异症发病中的作用。方法采用RT-PCR的方法检测35例卵巢子宫内膜异位囊肿患者的在位、异位内膜组织中紧密连接蛋白Claudin-4 mRNA的表达,并与30例正常对照子宫内膜比较。结果 ①Claudin-4 mRNA在在位、异位及对照组织中表达量分别为12.38±6.12,4.59±1.35和13.05±6.48。异位内膜组织中Claudin-4 mRNA的表达量低于在位和对照组内膜组织,差异有统计学意义(P〈0.01)。在位与对照内膜组织比较,Claudin-4mRNA的表达量差异无统计学意义(P〉0.05)。②在异位内膜组织中,Ⅰ,Ⅱ期患者的Claudin-4 mRNA的表达量(6.74±1.92)较Ⅲ,Ⅳ期患者(3.82±1.01)明显升高,差异也有统计学意义(P〈0.01)。③在位与对照内膜组织中,增生期Claudin-4 mRNA的表达量(14.01±7.02)较分泌期(11.14±5.48)明显升高,差异也有统计学意义(P〈0.05); 异位组内膜组织中,增生期与分泌期Claudin-4 mRNA的表达量差异无统计学意义(P〉0.05)。结论 Claudin-4在异位内膜中低表达可能与子宫内膜异位症的发生发展有关。
Objective To investigate mRNA expression of claudin 4 in the eutopic and ectopic endometrium of women with endometriosis and normal endometrium,and evaluate the role of clandin-4 in the pathogenesis of endometriosis. Methods The mRNA expression of elaudin-4 in eutopie and eetopic endometrium of patients with endometriosis (n= 35) and in normal endometrium from control group (n=30) were detected by reverse transcription-polymerase chain reaction. Results ① In eutopic and ectopic endometriam of women with endometriosis and control endometrium,the expression of claudin-4 mRNA was 12.38±6.12,4.59±1.35 and 13.05±6.48, respectively. Expression o{ claudin-4 mRNA was significantly lower in the eetopic endometriotic tissue than in the eutopic endometrium and the control endometrium (P〈 0.01). No significant difference was found Between eutopic endometrium from women with endometriosis and control endometrium (P〉0.05). ② Expression of claudin-4 mRNA levels of mild endometriosis were significantly higher than that of severe endometriosis in the ectopic endometriotic tissue ( 6.74±1.92 vs 3.82±1.01 ) (P〈 0.01 ). ③ In eutopic and control endometriam, Expression of claudin-4 mRNA levels were significantly higher in the proliferative phase than that in the secretory phase (14.01 ±7.02 vs 11.14 ± 5.48)(P〈0.05). In eetopie endometriam, no significant difference was found between proliferative phase and secretory phase (P〉0.05). Conclusion Down-regulated expression of claudin-4 could be involved in the development of endome-triosis lesions.
