摘要
目的分析体检健康人群血清大豆蛋白特异性Ig G(s Ig G)抗体水平,探讨其与年龄、性别的关系,并分析抗体识别蛋白质组分。方法丙酮溶解法制备大豆蛋白粗提液,包被微孔板,ELISA法检测240例体检健康者血清大豆蛋白s Ig G抗体水平,分析抗体水平与年龄、性别的关系;十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)分析大豆蛋白质组分,并用酶免疫印迹技术分析与Ig G特异性结合的主要蛋白质组分。结果体检人群大豆蛋白s Ig G抗体阳性率为55.8%,女性(66.6%)显著高于男性(45.0%),差异有统计学意义(P<0.01)。男性各年龄组间s Ig G抗体阳性率差异有统计学意义(P<0.01),且男性组的阳性率随年龄增高呈上升趋势;酶免疫印迹技术分析结果显示,不同个体间可能存在针对不同大豆蛋白抗原组分的s Ig G抗体。结论体检健康人群中普遍存在针对大豆蛋白质组分的s Ig G抗体,且s Ig G抗体水平与年龄和性别相关。
Objective To detect the levels of soy protein-specific Ig G antibody in healthy examination population,explore its correlations with age and gender and analyze the components recognized by the specific antibody. Methods The soy proteins were extracted with the acetone extraction method,and the crude proteins were coated on the microtiter plates. The levels of serum soy protein-specific Ig G antibody in 240 health examination subjects were detected with enzyme-linked immunosorbent assay( ELISA). Te correlations of the antibody levels with age and gender of the subjects were analyzed. The components of soy proteins were analyzed by SDS-PAGE,and the main components recognized by the soy protein-specific Ig G antibody were identified by western blot. Results The positive rate of soy protein-specific Ig G antibody in healthy examination population was 55. 8%,which in females was significantly higher than that in males( 66. 6% vs 45. 0%,P〈0. 01). In the males the positive rates of specific Ig G antibody showed statistically significant different between each age group( P〈0. 01),and the positive rate of male group increased with age on the rise. western blot showed that there were distinct specific Ig G antibodies to different components of soybean protein components in various individuals. Conclusion There are soy protein-specific Ig G antibodies in general population,and the levels of the antibodies may be related to age and gender.
