密蒙花总黄酮的抗氧化及免疫调节作用 被引量:5

摘要 为研究密蒙花总黄酮(TFB)对D-半乳糖所致衰老小鼠的抗氧化和免疫调节作用,以200 mg/(kg·d)的剂量对昆明雄性小鼠皮下注射D-半乳糖建立衰老模型小鼠,对模型小鼠分别灌胃TFB 25、50、100 mg/(kg·d)或维生素E 50 mg/(kg·d),正常对照组及模型对照组灌服等剂量的蒸馏水,连续灌胃40 d,取血清测定谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)、乳酸脱氢酶(LDH)及总胆红素(TBIL)和总蛋白(TP)含量,取肝脏与脾脏计算脏器指数并测定肝及脑组织中超氧化物歧化酶(SOD)活力及谷胱甘肽(GSH)和丙二醛(MDA)含量,并做组织切片观察肝脏组织病理学变化。结果表明,与模型对照组比较,50、100 mg/(kg·d)剂量的TFB能显著抑制衰老小鼠血清AST、AST、ALP、LDH活力和TBIL含量的升高;并且能使小鼠肝和脑组织中的SOD活力和GSH含量显著升高,使MDA含量降低。其中,50 mg/(kg·d)剂量组使肝和脑组织中SOD活力分别显著升高10.49%和26.16%,使GSH含量分别显著升高42.10%和31.64%,使MDA含量分别显著降低55.03%和20.86%。各剂量组TFB均能明显提高衰老小鼠的脾脏指数和胸腺指数,50 mg/(kg·d)TFB剂量组使胸腺指数显著升高23.08%。肝组织切片病理学检查显示,TFB各剂量均可以减轻肝脏组织病理性改变。综上,TFB对D-半乳糖损伤模型小鼠的损伤具有改善作用。 To investigate the regulatory effects of total flavonoids from Buddleja officinalis Maxim DC.( TFB) on the immune function and antioxidant mechanism in aged mice induced by D-galactose,mice were subcutaneously injected with D-galactose at a dosage of 200 mg / kg body weight once daily to make aged model. Experiment groups were fed with TFB at the dose of 25,50,100 mg /( kg·d) and vitamin E50 mg /( kg·d) by gavage,respectively; the normal control group and the model control group were fed with the same amount of distilled water once daily,and the experiment lasted 40 days. The activities of alkaline phosphatase( ALP),glutamate pyruvate transaminase( ALT),aspartate aminotransferase( AST),lactate dehydrogenase( LDH),total bilirubin( TIBL) and total protein( TP) level in serum,and the levels of malondialdehyde( MDA),glutathione( GSH) and the activities of superoxide dismutase( SOD) in the liver and brain tissue were assayed using standard procedures. Immune function in aged mice were used to detect the thymus gland index and spleen index. Meanwhile hepatic histopathological examination wasobserved. The results showed that TFB at 50,100 mg /( kg · d) investigated doses significantly lowered the levels of serum ALT,AST,ALP,LDH,TIBL,enhanced the activities of SOD,increased the level of GSH,and lowered the levels of MDA of liver and brain. Especially,TFB at 50 mg /( kg·d) investigated doses significantly enhanced the activities of SOD by 10. 49% and 26. 16%,increased the level of GSH by 42. 10% and 31. 64%,lowered the level of MDA by 55. 03% and 20. 86% in liver and brain,respectively. The thymus gland index and spleen index were all higher in the TFB dose groups,TFB 50 mg /( kg· d) increased the thymus gland index by 23. 08%. Meanwhile,ameliorating the liver damage were observed in the TFB groups in mice. Thus,TFB was able to enhance the immune function and improve the antioxidant capacities in aged mice induced by D-galactose.
