摘要 利用亚麻刺盘孢(Colletotrichum lini ST-1)静息细胞转化去氢表雄酮,制备三羟基雄甾烯酮,通过单因素优化确定了最佳摇瓶转化条件:细胞质量浓度为12 g/L,p H值为6.5,装液量为30 m L/250 m L,转速为220 r/min,温度为30℃。在上述条件下,底物投料质量浓度为10 g/L时,转化48 h,产物摩尔得率为38.5%,较生长细胞提高了21.0%。在上述工作基础上,尝试了静息细胞连续批次转化,最终通过两批次的连续转化,在底物累计投料质量浓度为18 g/L时,转化78h,产物的累积质量浓度高达12.1 g/L,产物摩尔得率可达60.5%。 The bioconversion of dehydroepiandrosterone (DHEA) to 3 (3,7a , 15a - trihydroxy-5- androsten-17-one by Colletotrichum lini ST-1 resting cells was investigated. The optimal transformation parameters in flasks were cell concentration 12 g/ L,pH 6.5, liquid volume 30 mL/250 mL, rotational speed 220 r/min and temperature 30℃ , which resulted in a molar product yield of 38.5% for 7a , 15a -diOH-DHEA that was 20.1% higher than that from growing cells. Based on these conditions, two-batch bioconversion for 78 li with resting cells at a total substrate concentration of 18 g/L resulted in a concentration of 12.1 g/L and a molar product yield of 60.5% for 7a , 15a - diOH-DHEA.
