摘要 为探索竹荪多糖(DI)对巨噬细胞RAW264.7免疫功能的影响,分别采用四唑氮蓝(MTT)法、中性红法、Griess法、ELISA法,检测DI对巨噬细胞增殖、吞噬活性、一氧化氮(NO)和白细胞介素1(IL-1)、白细胞介素6(IL-6)、α肿瘤坏死因子(TNF-α)分泌量的影响;运用反转录PCR(RT-PCR)检测i NOS和IL-1、IL-6、TNF-α的m RNA表达量的变化。结果表明,在25-200μg/m L质量浓度范围内,DI能够明显促进巨噬细胞增殖,增强其吞噬活性。同时,DI能够增强巨噬细胞分泌NO、IL-1、IL-6、TNF-α的能力,且明显提高i NOS、IL-1、IL-6、TNF-αm RNA的表达水平。因此,竹荪多糖对巨噬细胞RAW264.7具有免疫刺激作用。 In order to investigate the immunomodulatory effects of a polysaccharide from Dictyophora indusiata (DI) on the immune function of RAW264.7 cells,the effects of DI on cell proliferation,phagocytosis ofRAW264.7 cells,levels of NO,/ L-1 ,/ L-6 and TNF-a production were detected by MTT assay,neutral red test,Griess method,and ELISA, respectively. The mRNA expression level of iNOS, IL-1 , IL-6 and TNF-a were also measured by reverse transcription polymerase chain reaction (RT- PCR). The results indicated that DI contributed significantly to the proliferation of the macrophages and the phagocytic activity was at 25-200 jjug / mL. In addition, the production of NO,/ L-1 , IL-6 and TNF-a in macrophages were increased. Further,the expression of iN0S , IL-1 , IL-6 and TNF-a mRNA were significantly upregulated. The DI played immunomodulatory effects on macrophage cell line RAW264.7.
