蒽贝素对大鼠胶原诱导性关节炎炎症评分及炎症细胞因子的影响

Effects of Embelin on Arthritis Score and Inflammatory Cytokines in Rats with Collagen-Induced Arthritis

目的探讨蒽贝素对胶原诱导性关节炎(collagen induced arthritis,CIA)大鼠炎症评分及炎症细胞因子的影响。方法将50只大鼠随机分为正常对照组(8只)和造模组(42只)。造模组大鼠采用注射牛Ⅱ型胶原(bovine typeⅡcollagen,CⅡ)+完全弗氏佐剂(complete Freund’s adjuvant,CFA)乳剂进行初次免疫,1周后注射CⅡ+不完全弗氏佐剂(incomplete Freund’s adjuvant,IFA)乳剂加强免疫,建立CIA模型。将造模成功的32只CIA大鼠随机分为4组:CIA模型组、甲氨蝶呤组(1 mL·100g^(-1)灌胃,2次·周-1)和蒽贝素低剂量、高剂量2组(均以1mL·100g^(-1)灌胃,1次·d-1),每组8只。CIA模型组、正常对照组均以0.5%羧甲基纤维素钠1mL·100g^(-1)灌胃,1次·d-1。分别于实验第14、21、28、35天各组称体质量及对关节炎症进行评分。实验第36天检测各组血清肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1β(interleukine-1β,IL-1β)的水平。结果与正常对照组比较,CIA模型组、甲氨蝶呤组,蒽贝素低剂量、高剂量2组实验第14、21、28、35天体质量均减轻和关节炎症评分及实验第36天血清TNF-α、IL-1β水平均明显升高(均P<0.05);与CIA模型组比较,甲氨蝶呤组,蒽贝素低剂量、高剂量2组实验第21、28、35天体质量均增加和关节炎症评分及实验第36天血清TNF-α、IL-1β水平均明显降低(均P<0.05);与甲氨蝶呤组比较,蒽贝素高剂量组实验第28、35天体质量均增加,实验第36天血清IL-1β含量明显降低(均P<0.05)。结论蒽贝素可降低胶原诱导性关节炎大鼠炎症细胞因子水平,缓解关节炎的症状,具有抗炎作用,可能对类风湿关节炎有一定的疗效。

Objective To investigate the effects embelin on arthiritis score and inflammatory cytokines in rats with collageninduced arthritis（CIA）.Methods Fifty rats were randomly divided into normal control group（n=8）and model group（n=42）.The CIA was induced by the first immunization with type Ⅱ collagen（CⅡ）in complete Freund’s adjuvant（CFA）and the second immunization with CⅡ in incomplete Freund’s adjuvant（IFA）after 1 week.Then,32 CIA rats were randomly divided into four groups：CIA model group,methotrexate group（1 mL·100 g^-1 intragastric administration,twice per week）,low dose of embelin group（1 mL·100 g^-1 intragastric administration,once per day）,and high dose of embelin group（1 mL·100 g^-1 intragastric administration,once per day）,with 8 rats in each group.Rats in normal control group and CIA model group were given intragastric administration of 0.5% sodium carboxymethyl cellulose 1 mL·100 g^-1 once per day.Body weight and arthiritis score were measured on days 14,21,28 and 35 of the experiment.Serum concentrations of tumor necrosis factor-α（TNF-α）and interleukine-1β（IL-1β）were detected on day 36 of the experiment.Results Compared with normal control group,body weight decreased and arthiritis score increased on days 14,21,28 and 35,and serum concentrations of TNF-αand IL-1βincreased on day 36 in CIA model group,methotrexate group,embelin group, low dose of embelin group and high dose of embelin group（P 〈0.05）.Compared with CIA model group,body weight increased and arthiritis score decreased on days 21,28 and 35,and serum con-centrations of TNF-α and IL-1β decreased on day 36 in methotrexate group,embelin group,low dose of embelin group and high dose of embelin group（P 〈0.05）.Compared with methotrexate group,body weight increased on days 28 and 35 and serum concentrations of IL-1β decreased on day 36 in high dose of embelin group（P 〈0.05）.Conclusion Embelin can decrease the levels of inflammatory cytokines and relieve the symptoms of arthritis in CIA rats,suggesting that embelin may be effective for rheumatoid arthritis.
