摘要
目的:通过高脂饲料诱发的SD雄性大鼠肥胖模型研究肥胖对雄性生殖和后代代谢的损伤作用。方法:通过喂养高脂饲料建立雄性大鼠肥胖模型;精子质量检测系统检测父代大鼠精子活动能力;子代25周龄时采用糖耐量实验和胰岛素耐量实验检测糖代谢情况;HE染色检测胰腺形态;结果:高脂饲料喂养雄性大鼠5周后出现明显肥胖,且精子活力明显下降;相比于子代对照组,子代实验组大鼠糖耐量和胰岛素耐量均出现受损趋势;子代实验组大鼠胰岛面积变小。结论:肥胖能够损伤雄性大鼠的精子活力,可能是后代成年期发生代谢性疾病的高风险因素。
Objective:To investigate the influence of high-fat diet induced obesity on rat sperm motility and metabolism in offspring.Method:Obesity rat models were established by high-fat diet feeding.Sperm quality detection system was performed to analyze motility of sperm.Glucose and insulin tolerance tests(GTT and ITT)were performed in 25 weeks age of offspring.Pancreatic morphology was analyzed by HE staining.Result:Compared with normal diet rat group,high-fat diet rat group tended to be obesity obviously five week later.And obviously,obesity rat had sperm motility impairment.Compared with offspring control group,offspring experiment group showed more severe impairment in GTT,ITT and islet area.Conclusion:High-fat diet causes sperm motility impairment in male rats,and may be a high-risk factor of metabolism disease in offspring.
