摘要
目的合成一种对动脉粥样硬化病灶靶向的纳米MRI造影剂。方法将3-氨丙基三乙氧基硅烷(APTES)包裹到四氧化三铁纳米粒的表面,再将具有动脉粥样硬化靶向性的Oligo适配子与纳米粒偶联。通过XRD、IR、TEM等分析手段对硅烷修饰前后磁性纳米粒进行表征.并用HPLC检测反应上清液DNA含量,以评估适配子与纳米粒偶联程度。结果硅烷成功包裹到纳米粒上,包裹后的纳米粒粒径为30 nm,为反尖晶石结构,且适配子与纳米粒成功偶联,偶联后粒径为170 nm。结论成功制备出动脉粥样硬化靶向纳米MRI造影剂,方法稳定,操作便捷。
OBJECTIVE To synthesize a new target of Manometer MRI contrast agents as a MRI contrast agent in atherosclerosis. METHODS Using the APTES coated magnetite nanoparticles and atherosclerosis target-based Oligo aptamer was cross-linked with particles. The nanoparticles were characterized by XRD, IR and TEM, and using HPLC to detect that DNA content of supernatant fluid to identify the connection rate. RESULTS The APTES coated magnetite nanoparticles successfully and the particle size of nanoparticles was 30 nm, and their morphology was inverse spinel, aptamer and nanoparticles success coupling. The particle size of the aptnanoparticles was 170 rim. CONCLUSION Synthesized a new target of Manometer MRI contrast agents successfully, the method is stable and the operation is very convenient.
