“标本配穴”电针和限食干预对肥胖大鼠褐色脂肪组织SIRT1和UCP1的影响 被引量:8

摘要 目的:研究"标本配穴"电针和饮食控制对肥胖大鼠褐色脂肪组织沉默信息调节因子1(silent information regulator1,SIRT1)和解偶联蛋白(uncoupling protein,UCP1)的影响,比较其单独及协同作用.方法:将40只高脂喂养的肥胖大鼠随机分为4组,每组10只.模型对照组(C)继续高脂喂养,电针组(E)给予"标本配穴"电针治疗,限食组(R)每只给予对照组摄食量的70%,电针+限食组(E+R)给予"标本配穴"电针结合限食干预治疗,共电针及限食8 wk.检测各组大鼠体质量和血清葡萄糖(fasting plasma glucose,FPG)、胰岛素(fasting insulin,FINS)、甘油三酯(triglyeride,TG)、总胆固醇(total cholesterol,TC)水平变化,对褐色脂肪组织进行SIRT1蛋白和UCP1m RNA的测定.结果:与C组比较,3个电针和饮食干预组体质量及FPG、FINS、TG和TC均明显下降(P<0.01),且E+R组下降程度优于单独电针或限食组(P<0.05);与C组比较,3个干预组褐脂中SIRT1蛋白表达水平明显升高(P<0.01);与C组比较,3个干预组褐脂中UCP1 m RNA表达明显升高(P<0.01),且E+R组效果明显优于单独电针或单独饮食干预组(P<0.05).结论:"标本配穴"电针和限食可能是通过促进褐色脂肪组织中SIRT1和UCP1的表达,增加产热而发挥减肥作用."标本配穴"电针法抵抗肥胖的功效与饮食控制相当,二者之间存在一定的协同作用. AIM: To observe the effect of "Biao-Ben points association" electro-acupuncture combined with dietary control on SIRT1 and UCP1 expression in brown adipose tissue of obese rats.METHODS: Forty rats with obesity induced with a high-fat diet were randomly divided into four groups, with 10 rats in each group. Rats in the model control group(C) were continued to be given a high-fat diet. Rats in the electroacupuncture group(E) were treated with "BiaoBen points association" electro-acupuncture. The dietary restriction group(R) was fed only 70% of high-fat diet to the control group. The electro-acupuncture combined with dietary restriction group(E + R) was treated with both acupuncture and dietary restriction. After 8 wk of treatment, changes of body weight, fasting glucose, insulin, triglycerides, and total cholesterol were recorded. The expression of silent information regulator 1(SIRT1) and uncoupling protein 1(UCP1) m RNA was detected in brown adipose tissue.RESULTS: Compared with the C group, body weight, FPG, FINS, TG, and TC in the otherthree intervention groups were significantly lower(P〈0.01); these indexes were also significantly lower in the E + R group than in the other two intervention groups(P〈0.05). Compared with the C group, the expression of SIRT1 in the other three intervention groups was significantly higher(P〈0.01). Compared with the C group, the expression of UPC1 m RNA in the other three intervention groups was significantly higher(P〈0.01); UPC1 m RNA expression in the E + R group was also significantly higher than that in the other two intervention groups(P〈0.05).CONCLUSION: "Biao-Ben points association" electro-acupuncture and dietary restriction might increase the heat production and result in weight loss by promoting the expression of SIRT1 and UCP1 in brown adipose tissue. The anti-obesity effect of "Biao-Ben points association" electro-acupuncture is comparable to that of dietary restriction, and there exist some synergistic effects between them.
