miR-140-5p调控FGF9在拇外翻中的作用

摘要 目的探讨miR-140—5p调控FGF9在拇外翻中的作用,从基因层面阐明拇外翻的发病机制,为临床预防和治疗拇外翻提供基础理论依据。方法收集2013—2015年于抚顺市中心医院就诊的拇外翻患者(10例)和非拇外翻患者(5例)的第一跖骨内侧骨组织及表面软组织样本,运用Western blot以及real—timePCR检测各组织中miR-140—5p和FGF9的表达.并对其表达差异进行分析。利用人成骨细胞系hFOBI.19对骨保护素(OPG)和骨钙素(OCN)进行Western blot分析,验证miR-140—5p和FGF9的调控关系和对成骨细胞成骨能力的影响。结果拇外翻患者第一跖趾关节与非拇外翻患者比较,miR-140—5p表达显著下降(P=0.0065),FGF9表达明显增高(P=0.0055)。上调miR-140—5p后FGF9mRNA表达显著下降(P=0.0050),而下调miR-140—5p后FGF9mRNA表达显著增加(P=0.0035)。与未转染组比较上调miR-140—5p后OPG(P=0.0045)和OCN蛋白(P=0.0037)表达明显增强,而下调miR-140—5p后OPG(P=0.0047)和OCN蛋白(P=0.0049)表达减弱。结论本研究初步证明了miR-140—5p与FGF9在拇外翻的病理过程中具有重要作用,这为拇外翻的病因以及诊治研究提供了新的方向。 Objective To discuss effects of miR-140-Sp on FGF9 in hallux valgus, illuminate the pathogenesis of hallux valgus from the point of gene, which provides basic theoretical foundation for the treatment and prevention of hallux valgus. Methods Samples of patients with hallux valgus (10 cases) and without hallux valgus (5 cases) in the Fushun Central Hospital from 2013 to 2015 were collected, thus the expression of miR-140-Sp and FGF9 in the first metatarsal medial bone tissues and the surface of the soft tissues were analyzed with Western blot and real-time PCR. Besides, Western blot analysis on OPG and OCN with osteoblast cell line hFOBI.19 were conducted; the effects and regulation relationship between miR-140-Sp and FGF9 on the osteogenic capacity of osteoblast cell line were verified. Results Compared with the first plantar toe joint of the patients without hallux valgus, the miR-140-Sp of hallux val- gus patients reduced remarkably (P = 0.0065), FGF9 mRNA increased obviously (P = 0.0055). After the increasing of miR-140-Sp, FGF9 mRNA was reduced remarkably (P = 0.0050), and after the reducing of miR-140-5p, FGF9 mR- NA was increased remarkably (P = 0.0035). Compared with the untransfected group, OPG (P = 0.0045) and OCN (P = 0.0037) protein increased after miR-140-Sp was increased. While the decrease of miR-140-Sp caused the weaken of OPG (P = 0.0047) and OCN protein (P = 0.0049). Conclusion This experiment primarily proves the important functions that miR-140-Sp and FGF9 having in the pathological process of hallux valgus, which provides new direction for the pathogenesis and diagnosis of hallux valgus.
