大鼠原位肝移植后受体源肝细胞产生情况的实验研究

摘要 目的:利用Y染色体检测经性别错配原位肝移植术后的SD大鼠受体移植肝组织中是否有受体源肝细胞产生。方法采用双袖套技术对SD大鼠行性别错配(雌鼠为供体,雄鼠为受体)肝移植,将造模成功的大鼠分为三组,分别于术后5、10、15 d将其处死(分别为A、B、C组,另设雌鼠为D组,雄鼠为E组),取其肝组织通过聚合酶链式反应( PCR)及琼脂糖凝胶电泳技术检测移植肝中是否存在雄性大鼠 Y 染色体特有的 SRY 基因,以明确经肝移植后移植肝中有无受体源肝细胞产生。结果三组大鼠受体移植肝中均未检测到 SRY 基因存在。结论 SD大鼠原位肝移植后移植肝短期内无受体源肝细胞产生。 Objective Using Y chromosome as natural maker to detect whether or not there was receptor-derived hepatocyte exist after sex-mismatched( female as donator,male as receptor)orthotopic liver transplantation of SD rats. Methods Orthotopic liver transplantation was performed by two-cuff method. The rats were executed after transplantation 5 days( group A),10 days( group B)and 15 days (group C),while female and male rats as the control group(group D,group E). Polymerse chain reac-tion( PCR)and agarose gel electrophoresis were used to detect the SYR gene,which was a specific marker of Y chromosome of male to prove the exist of receptor-derived hepatocyte. Results The SRY gene was not detected in all groups except group E. Conclusions The SD rats after orthotopic liver transplantation has no receptor-derived exist.
