摘要
目的:采用HPLC法同时测定天麻饮片中天麻素、对羟基苯甲醇、香荚兰醇、巴利森苷A、巴利森苷B、巴利森苷C等6种成分的含量。方法:采用Agilent 1220型高效液相色谱仪;Hypersil C_(18)色谱柱(250 mm×4.6 mm,5μm);流动相为0.5%醋酸水溶液(A)-0.5%醋酸甲醇溶液(B),洗脱梯度:0~10 min,98%A,10~60 min,98%~60%A,60~75 min,60%A;体积流量为0.8 m L·min^(-1);检测波长为270 nm;柱温为30℃;进样量为20μL。结果:6种成分线性关系良好(r≥0.999 3);平均加样回收率在96.99%~101.73%,RSD<2.5%。29批天麻饮片的测定结果表明,云南与四川地区天麻素含量最高。6种成分因地区不同,含量有所差异,其中巴利森苷A类含量最高,对羟基苯甲醇和香荚兰醇含量较低。结论:建立的方法快速简便、稳定可靠,能更全面、有效地对天麻饮片进行质量控制。
Objective: To determine 6 active components (gastrodin, 4-hydroxybenzyl alcohol, vanillyl alcohol, parishin A, parishin B, parishin C) of Gastrodiae Rhizomaby HPLC. Methods: The separation was carried out by an Hypersil C18 column (250 mm x 4. 6, 5 Ixm) in a mobile phase consisting of 0. 5% CH3COOH - H2O(A) and 0.5% CH3COOH - CH3OH(B) with a gradient elution system (0→10 min, 98%A; 10→60 min, 98%→60%A; and 60→75 min, 60%A). Detection wavelength was set at 270 nm and column temperature was 30℃. Injection volume was 20 IxL. Results: The six components had good linearity ( r≥0. 999 3 ). The average recovery rates were 96. 99% - 101.73% with RSD 〈 2. 5%. Gastrodiae Rhizoma of 29 different batches was determined, in which Gastrodiae Rhizoma from Yunnan and Sichuan areas had the highest content of gastrodin. The contents of 6 components in Gastrodiae Rhizoma varied in different areas. The contents of parishin A was the highest while the contents of 4-hydroxybenzyl alcohol and vanillyl alcohol were lower. Conclusion: The method is rapid, simple, accurate and can be used to control quality of Gastrodiae Rhizoma in a more comprehensive and effective way.
