摘要
MYB转录因子参与植物对外界逆境胁迫反应的调控,为提高水稻的耐盐性,本研究对水稻OsMYB56基因进行了生物信息学分析,利用RT-PCR技术克隆了水稻耐盐碱基因OsMYB56,构建了植物表达载体pCAMBIA3300-Os MYB56,并通过农杆菌介导法将OsMYB56基因导入水稻中,以进一步研究其功能。对转化植株进行PCR、RT-PCR、Southern杂交及Bar蛋白试纸条检测,证明OsMYB56基因已整合到水稻基因组中。本研究所获得的转基因水稻材料对水稻耐盐碱育种具有重要的意义。
MYB transcription factors are related to salt tolerance of plant. In order to improve salt tolerance of rice, in this study, the OsMYB56 gene was cloned by RT-PCR technique and the plant expression vector pCAMBIA3300- OsMYB56 was constructed, OsMYB56 gene was successfully transformed into super rice 'Jijing 88' mediated with Agrobacterium. The transgenic plants were confirmed by PCR, RT-PCR, Southern blot and Bar quick strip. The transgenic plants can be employed as important materials in rice breeding for salt tolerance.
