摘要
目的:分析地肤病毒病的病原。方法:在生物学检测的基础上,提取病毒双链RNA(dsRNA),合成单链cDNA后经多重PCR扩增、测定序列并分析,对地肤病毒病进行了鉴定。结果:有明显花叶症状的地肤由蚕豆萎蔫病毒2号(Broad bean wilt virus 2,BBWV2)侵染所致。将地肤分离物(Isolate-DF)中部分病毒序列进行比对分析,发现其与已报道的15个不同来源BBWV2序列的同源性为79.9%~88.0%。结论:通过分子鉴定,首次证实我国药用植物地肤上发生的花叶病毒病系由BBWV2侵染所致。
Objective:Analyze the pathogeny on the Kochia scoparia virus disease. Method:The Kochia scoparia virus was identified,the virus double-stranded RNA (dsRNA)was extracted,and the single cDNA was synthesized using the multiplex PCR amplification method,and the target sequence was analyzed. Result:The Mosaic symptoms of K. scoparia were caused by the infection of Broad bean wilt virus 2 (Broad bean wilt virus 2,BBWV2). Sequence homology analysis revealed that the partial virus sequence from Isolate-DF of K. scopria showed 79.9%~88.0% homology with other 15 reported BBWV sequences. Conclusion:The results of nucleotides homology analyzing firstly confirmed that the Mosaic virus disease of medicinal plant K.scopria was caused by BBWV2 in china.
