摘要
目的对颈总动脉穿刺法及传统线栓法制备大鼠大脑中动脉栓塞(middle cerebral artery occlusion,MCAO)模型的时效性、成功率、死亡率比较。方法将32只雄性大鼠随机分为颈总动脉穿刺法组与传统线栓法组,将线栓从颈总动脉插入,栓塞大脑中动脉。采用Bederson神经功能评分标准,于术后24 h进行神经功能评分,并统计24 h成功率及死亡率。结果颈总动脉穿刺法和传统手术方法制备大鼠MCAO模型的插线栓时间、成功率、死亡率分别为(82.3±17.4)s和(164.6±22.0)s,差异有显著性(P<0.01);87.5%和68.75%,差异无显著性(P>0.05);6.25%和18.75%,差异无显著性(P>0.05)。结论颈总动脉穿刺法制备大鼠大脑中动脉栓塞模型简单省时,创伤小,可操作性强。
Objective To compare the timeliness,success and mortality rates between the modified carotid artery puncture method( MCAPM) and standard suture method( SSM) in the establishment of rat model of a middle cerebral artery occlusion( MCAO). Methods Thirty-two male rats were randomly and equally assigned into MCAPM group and SSM group. The MCAO models were established by inserting a thread into the common carotid artery( CCA). 24 h after modeling,the rats of the two groups were evaluated with Bederson neurological scores,and the modeling success rate and mortality rate were analyzed. Results The suture insertion times,success rates and mortality rates of the MCAPM vs.SSM groups were( 82. 3 ± 17. 4) s versus( 164. 6 ± 22. 0) s( P 〈 0. 01),87. 5% versus 68. 75%( P 〉 0. 05),and6. 25% versus 18. 75%( P 〉 0. 05). Conclusions MCAPM can be used to establish the rat model of MCAO due to its simplicity,mild wound and feasibility.
