醇提与水提工艺消栓通络片对大鼠局灶性脑缺血的治疗作用对比

A comparison study of the difference in effect between adjusted Xiao Shuan Tong Luo and marketed Xiao Shuan Tong Luo Tablets on rats with focal cerebral ischemia

摘要 目的对比研究改工艺后消栓通络片与市售消栓通络片对大鼠局灶性脑缺血的治疗作用差异。方法采用颈内动脉插入尼龙线栓的方法建立大鼠左侧大脑中动脉栓塞(middle cerebral artery occlusion,MCAO)模型。观察给药后各组大鼠行为学评分、测定脑梗死面积及血清超氧化物歧化酶(SOD)、NF-κB、caspase-3含量的差异。结果改工艺后的消栓通络片高、中剂量组均能有效缓解4h,24h时MCAO引起的行为学病变,降低行为学评分;高、中、低剂量组均能升高MCAO引起的SOD降低;高、低剂量组能明显降低MCAO引起的血清NF-κB的升高;中、低剂量组对MCAO引起的Caspase-3的显著升高有一定抑制作用。与市售消栓通络片比较,改工艺消栓通络片从大鼠行为学评分的降低,血清SOD活性的增加,血清Caspase-3、NF-κB的表达效果均有优于市售消栓通络片的趋势。结论改工艺消栓通络片各剂量组对脑缺血进展期大鼠均有一定的恢复作用,其药效略优于市售消栓通络片。 Objective To explore the difference in effect between adjusted Xiao Shuan Tong Luo and marketed Xiao Shuan Tong Luo tablets on rats with focal cerebral ischemia. Methods Rat models with middle cerebral artery occlusion( MCAO) were established. The behavior,infarction size,SOD activity,NF-κB and CASPASE-3 contents were measured. Results Compared with the lower dosage group,higher and middle dosage groups of Xiao Shuan Tong Luo( 0. 44 and 0. 22 g·kg^- 1) could remarkably decrease the scales of neurological behavior of MCAO rats. All group of Xiao Shuan Tong Luo could obviously increase the SOD of MCAO rats remarkably,and higher and lower dosage groups of Xiao Shuan Tong Luo could obviously decrease the NF-κB lever of MCAO rats. Middle and lower dosage groups of Xiao Shuan Tong Luo could obviously decrease the CASPASE-3 level of MCAO rats. Compared with marketed Xiao Shuan Tong Luo tablets,adjusted Xiao Shuan Tong Luo was prior in the decrease of behavior evaluation,increase in SOD,and the expressions of serum Caspase-3 and NF-κB. Conclusions Each dosage group of adjusted Xiao Shuan Tong Luo is helpful in the recovery of rats with cerebral ischemia,whose effect is better than marketed Xiao Shuan Tong Luo tablets.
