摘要
[目的]探讨续断提取物对血管性痴呆模型大鼠学习记忆能力的影响.[方法]取雄性SD大鼠,随机分为正常对照组、假手术组、模型对照组和续断提取物高、中、低剂量(13.5,4.5,1.5g/L)组.采用反复夹闭、再通双侧颈总动脉同时腹腔注射给予硝普钠方法建立大鼠血管性痴呆模型.模型建立成功后灌胃给予各给药组相应剂量的药物,每日1次,连续28d.利用Morris水迷宫试验检测各组大鼠学习记忆能力;取心脏血测定超氧化物歧化酶(SOD)、丙二醛(MDA)及谷胱甘肽过氧化物酶(GSH-Px)水平.[结果]与正常对照组和假手术组比较,模型对照组寻找站台潜伏期明显延长(P<0.05),撤出平台后穿越平台次数明显减少(P<0.05),血清中MDA含量明显升高(P<0.05),SOD及GSH-Px水平明显降低(P<0.05).与模型对照组比较,续断提取物中、高剂量组寻找站台潜伏期明显缩短,血清中MDA含量明显降低(P<0.05),SOD及GSH-Px水平明显升高(P<0.05),穿越平台次数明显增加(P<0.05).[结论]续断提取物可改善血管性痴呆模型大鼠学习记忆能力.
OBJECTIVE To investigate the effects of Dipsacus asper extract on learning and memory ability of rats with vascular dementia (VD). METHODS Male SD rats were randomly divided into normal group, sham-operation group, model group, high, middle and low dose of Dipsacus asper extract groups (13.5,4. 5,1.5 g/L). VD model in rats was successfully established by repeated occlusion and recanalization of bilateral common carotid artery combined with intraperitoneal injection of sodium nitroprusside. After the operation, the rats in the Dipsacus asper extract groups were kept on intragastric administration of Dipsacus asper extract per day for 28 days respectively. Morris water maze was used to detect the learning and memory ability of rats. The levels of SOD, MDA and GSH-Px in the heart blood were determined. RESULTS Compared with the normal and the sham-operation group, the latent period was obviously increased (P〈0.05), and the times of traversing terrace were significantly decreased after removing the terrace in the model group (P〈0.05), MDA content in blood was increased (P〈0.05) and the activity of SOD and GSH-Px were decreased(P〈.05) in the model group. Compared with the model group, the latent period was reduced in the middle and high dose of Dipsacus asper extract group ( P 〈 0.05), MDA content in blood was decreased (P〈0.05) and SOD and GSH-Px activity were increased ( P 〈0.05), and the times of traversing terrace were obviously increased after removing the terrace in the middle and high dose of Dipsacus asper extract group ( P 〈0. 05). CONCLUSION Dipsacus asper extract can improve the learning and memory ability of rats with VD.
