芪胶升白胶囊对S180小鼠抗肿瘤作用及相关机制研究 被引量:3

Antitumor effect and related mechanism study of Qijiaoshengbai capsule on S180 tumor-bearing mice

摘要 目的:探讨芪胶升白胶囊对S180瘤株的抑制作用及机制。方法:以S180荷瘤小鼠为动物模型,试验设模型组,环磷酰胺组,芪胶升白胶囊0.5g/kg、芪胶升白胶囊1.0g/kg、芪胶升白胶囊2.0g/kg剂量组共5组,每日测量各组小鼠体重,各组分别连续给药15天后,取荷瘤小鼠血液进行血细胞计数,取瘤体、脾脏和胸腺,计算抑瘤率、脾指数和胸腺指数。选用AnnexinⅤ-FITC/PI双染流式细胞术测定肿瘤细胞的凋亡,用Western-blot法检测肿瘤组织中p53、bcl-2的表达情况,用免疫组化法检测CD34及VEGF的表达情况。结果:芪胶升白胶囊1.0g/kg剂量组能明显抑制小鼠移植性S180肿瘤生长,环磷酰胺、芪胶升白胶囊0.5g/kg、芪胶升白胶囊1.0g/kg、芪胶升白胶囊2.0g/kg剂量组抑瘤率分别为44.32%、17.18%、22.12%和8.74%。芪胶升白胶囊2.0g/kg剂量组肿瘤细胞凋亡率较环磷酰胺组明显提高,各剂量组可下调抗凋亡基因p53、bcl-2的表达及降低调控微血管密度和生长的CD34和VEGF的蛋白水平。结论:芪胶升白胶囊在体内对S180瘤株的生长有抑制作用,其抗肿瘤活性可能与抑瘤基因bcl-2、p53表达及微血管密度调控蛋白CD34、VEGF的水平变化有关。 Objective: To observe the inhibiting effects and related mechanism of Qijiaoshengbai Capsule on S180 tumor bearing mice. Methods: Mouse S180 cells were inoculated subcutaneously into the right armpit for 0.25 ml per mouse as animal models. Seventy-five mice were ran- domly assigned into control group, Cyclophosph.amide group, Low dose Qijiaoshengbai Capsule group, Middle dose Qijiaoshengbai Capsule group and High dose Qijiaoshengbai Capsule group, ecach groups have a drug administration and body weight respectively for 15 consecutive days, take the blood of tumor-bearing mice for blood test, dissect the body and take the tumor, spleen and thymus out to calculate tumor inhi- bition rate and thymus index, spleen index. In the aspect of mechanism,.we applied Annexin V-FITC/PI flow cytometer method to detect the apoptosis rate of tumor cells, Western - blot method to detect the expression of p53 and bcl-2 in tumor tissue, and applied immunohistochemi- cal method to detect the expression of VEGF and CD34. Result: Qijiaoshengbai Capsule in Middle dose group could be inhibit the growth of transplanting S180 mice tumor better, the tumor inhibition rate of cyclophosphamide group,Qijiaoshengbai Capsule in low, middle, high dose group respectively are 44.32%, 17.18% ,22.12% and 8.74%. The tumor cell apoptosis rate of Qijiaoshengbai Capsule in high dose group is higher than in cyclophosphamide group, and Qijiaoshengbai Capsule in each dose group can decrease the expression of p53, bcl-2, CD34 and VEGF,especially in middle dose group. Conclusion: The Qijiaoshengbai Capsule has the effect of restrain the growth of S180 tumor in vivo. The antitumor activity may be concerned with the expression of tumor suppression gene P53, bel-2 and MVD regulatory proteins CD34 and VEGF.
