摘要
目的:研究柚皮苷对人宫颈癌Hela细胞体外增殖的影响及相关机制研究。方法:用不同浓度的柚皮苷处理体外培养人宫颈癌Hela细胞,采用MTT法检测处理24、48、72 h后Hela细胞的增殖情况,流式细胞仪测定细胞凋亡率,Hoechst 33258染色观察细胞凋亡情况,同时Western blot观察环氧合酶-2(COX-2)表达的相对量。结果:与对照组相比,柚皮苷低剂量组细胞抑制率无显著差异(P>0.05),柚皮苷中剂量组和高剂量组细胞抑制率显著增高(P<0.05);柚皮苷低剂量组细胞凋亡率无显著差异(P>0.05),柚皮苷中剂量组和高剂量组细胞凋亡率显著增高(P<0.05),同时柚皮苷中剂量和高剂量组可观察到明显的细胞凋亡现象;柚皮苷低剂量组细胞COX-2表达的相对量无显著差异(P>0.05),柚皮苷中剂量组和高剂量组细胞COX-2表达的相对量显著降低(P<0.05);结论:柚皮苷能抑制宫颈癌细胞生长,且随其浓度增加抑制作用增强,其机制可能与其抑制COX-2蛋白表达,促进细胞凋亡相关。
Objective: To study the effect of naringin on human cervical cancer He La cell line and its related mechanism. Methods: He La cells were cultured in vitro in the presence of different concentration naringin. The method of MTT was used to detect the growth of He La cell in 24 h,48,72 h. Apoptotic cells were observed by Hoechst 33258 staining. The protein expressions of COX-2 in SCL-12 cells were analyzed by Western blot. Results: The inhibition ratio of the cell growth in middle dose group and high dose group significantly higher than control group( P〈0. 05) and the inhibition ratio in low dose group was no significant change( P〉0. 05). The apoptosis ratio of the cell growth in middle dose group and high dose group significantly higher than control group( P〈0. 05) and the apoptosis ratio in low dose group was no significant change( P〉0. 05). The expression of COX-2 was significantly decreased in middle dose group and high dose group. Conclusion: Naringin plays a central role in the apoptosis of the human cervical cancer He La cell line,which may be related to the expression of COX-2.
