摘要
通过文献搜索,收集整理了影响猪生长和肉质性状的显著单核苷酸多态性(single nucleotide polymorphism,SNP)位点,基于飞行时间质谱技术对大约克(n=400)和长白(n=399)样本进行SNP快速分型,并分析了这些位点在大约克和长白群体内的基因型、等位基因频率、哈代-温伯格平衡状态等遗传学特征。结果表明:利用飞行时间质谱技术可对17个影响猪肉质和生长性状的显著SNP位点进行同时快速基因分型,平均个体检出率为96.35%;17个被检SNP位点中9个为多态位点,其它8个为只有一种基因型的单态位点;多态SNP位点中有6个处于哈代-温伯格平衡状态,其它3个严重偏离哈代-温伯格平衡。本研究收集了对猪生长和肉质性状具有显著效应的SNP位点,探讨了利用飞行质谱技术进行多位点SNP快速分型的可行性,为今后建立猪生长和肉质性状标记辅助选择、加快猪的遗传研究进展奠定基础。
In the study,we collected the single nucleotide polymorphism( SNP) loci related with growth and meat quality of pigs by searching published papers and public databases.Based on the time-of-flight mass spectrometry( TOF-MS) technique,we genotyped the samples of Yorkshire( n = 400) and Landrace( n = 399),and analyzed the genotype frequency,allele frequency,and Hardy-Weinberg Equilibrium testing of these SNPs.The results showed that using TOF-MS,we successfully genotyped 17 SNPs at the same time,and the average call rate of these SNPs was 96.35%.Nine of the 17 SNPs were polymorphic loci,and the other 8 were monomorphic loci,which had only one genotype in both Yorkshire and Landrace.Additionally,for the 9 polymorphic SNPs,6 SNPs were in the Hardy-Weinberg Equilibrium and the other 3 deviated from it significantly.This study collected the significant SNPs related with growth and meat quality,and verified the effectiveness of genotyping multiple SNPs simultaneously and rapidly using TOF-MS,which provided foundations for the marker assisted selection( MAS) and the improvement of genetic progress of pigs.
