摘要
目的观察肝细胞肝癌(HCC)患者外周血单个核细胞(PBMC)中程序性死亡蛋白1(PD-1)和配体以及γ干扰素(IFN-γ)水平的变化。方法采集15例早期HCC患者、13例进展期HCC患者和12例健康人(对照组)的外周血并分离PBMC。流式细胞术检测PBMC中PD-1、PD-L1和PD-L2的表达,ELISA检测血清中IFN-γ的水平,利用Pearson相关系数法和One-way ANOVA分析PD-1与IFN-γ的相关性。结果进展期HCC患者外周血CD3+T细胞和CD19+B细胞中PD-1、PD-L1和PD-L2的表达水平和血清IFN-γ水平显著高于健康人群和早期HCC患者,HCC患者外周血CD3+T细胞和CD19+B细胞PD-1、PD-L1表达水平与IFN-γ呈正相关。结论肝癌患者PBMC中PD-1、PD-L1和PD-L2分子表达增加,且PD-1、PD-L1与IFN-γ水平有关联。
Objective To investigate the changes of programmed death γ ( PD-γ ) and ligands, as well as interferon-γ (IFN-γ) in peripheral blood mononuclear cells (PBMCs) of patients with hepatocellular carcinoma (HCC). Methods The peripheral blood was collected from 15 early HCC patients, 13 progressive HCC patients and 12 healthy volunteers. PBMCs was isolated from the peripheral blood. The expressions of PD-1, PD-L! and PD-L2 in PBMCs were detected by flow cytometry; the serum level of IFN-y was determined by ELISA; the correlation of PD-1 and IFN-y was analyzed with Pearson's correlation and One-way ANOVA. Results The expression levels of PD-1, PD-L1 and PD-L2 in CD3 + T cells and CD19+ B cells and serum IFN-γ level in progressive HCC patients were significantly higher than those in the healthy group and early HCC patients. The expression levels of PD-1, PD-L1 and PD-L2 in the CD3 + T cells and CDI9 ~ B cells of the HCC patients were positively correlated with IFN-γ. Conclusion The expression levels of PD-1, PD-L1 and PD-L2 increase in the PBMCs of HCC patients; PD-1 and PD-L1 are correlated with IFN-y level.
