针刺足三里穴对端粒酶基因敲除小鼠海马P2X受体及其表达的影响 被引量:1

摘要 目的观察针刺足三里穴对端粒酶基因敲除小鼠海马P2X受体表达的影响。方法采用随机数字表法将端粒酶基因敲除的早衰模型纯合子小鼠分为空白组、手针组及电针组,每组各6只小鼠。3组小鼠均正常饲养,期间空白组不给予任何特殊处理,手针组及电针组小鼠分别给予手针或电针刺激足三里穴。于治疗7d后观察各组小鼠海马区P2X受体及cAMP应答元件结合蛋白(CREB)表达间差异。结果①手针组及电针组海马区P2X。相对表达量[分别为(0.67±0.05)和(O.70±O.13)]均较空白组(0.79±0.03)有下降趋势,但各组间差异均无统计学意义(P〉0.05)。②电针组及手针组海马区P2X,表达[分别为(1.00±0.04)和(1.07±0.02)]均较空白组(1.21±0.05)明显下降(P〈0.05),但电针组与手针组海马区P2X,表达组间差异无统计学意义(P〉0.05)。③电针组小鼠海马区CREB表达(O.67±O.07)较空白组(0.82±0.05)及手针组(0.86±0.03)明显下降(P〈0.05)。结论电针可抑制端粒酶基因敲除小鼠海马区P2X。受体及CREB表达,其相关信号通路在针刺治疗神经退行性疾病的作用机制中可能发挥重要作用。 Objective To observe the expression of P2X receptors in the hippocampus of telomerase gene knockout mice after acupuncturing the bilateral ST36 acupoints. Methods Eighteen telomerase gene knockout homozygous mice were randomly divided into a blank control group ( group A) , a manual acupuncture group ( group B) and an electric acupuncture group ( group C) , each of 6. Group A was not given any interven- tion, while groups B and C were provided with manual or electric acupuncture at point ST36 for 7 days. The ex- pression of P2X receptors and cAMP response element bound protein (CREB) in the hippocampus were ob- served and compared among the 3 groups. Results The expression of P2X4 in the hippocampus was not signif- icantly different among the 3 groups in groups B and C decreased significantly compared with group A, but there were no significant differences between group Bs and C. The expression of CREB in group C decreased signifi- cantly compared with groups A and B. Conclusion EA inhibits the expression of P2X4 receptor and CREB the hippocampus. The signaling pathway may play an important role in the mechanism of action of acupuncture in neurodegenerative diseases.
