柠檬黄对小鼠卵母细胞成熟、体外受精及早期胚胎发育的影响研究 被引量:2

摘要 目的探讨柠檬黄对小鼠卵母细胞发育潜能的影响。方法选择成年雌性ICR小鼠24只,并每次分别给予柠檬黄0.25 g/kg(低剂量组,6只)、0.50 g/kg(中剂量组,6只)、1.0 g/kg(高剂量组,6只)、生理盐水(对照组,6只)1天/次,连续给药7天,统计卵母细胞数、孤雌激活率、体外受精率、卵裂率及囊胚形成率,观察卵母细胞纺锤体和染色体的形态,评估柠檬黄对小鼠卵母细胞发育潜能的影响。结果柠檬黄处理组小鼠卵母细胞的孤雌激活率、体外受精率、卵裂率及囊胚形成率均低于对照组(P<0.05)。纺锤体形态不正常,呈筒状和染色体排列不规则,散乱。结论柠檬黄降低了小鼠卵母细胞的发育潜能。 Objective To explore the effect of tartrazine on the developmental potential of mice oocytes. Method The adult female ICR mice(n = 24) were treated with different dose of tartrazine: 0.25 g/kg(lowdose group, n = 6), 0.50 g/kg(medium-dose group, n = 6) and 1.0 g/kg(high-dose group, n = 6) and normal saline(control group, n = 6) everyday uninterruptedly for 7 days. Then the mice were sacrificed to collect ovulated oocytes, and counted the parthenogenetic activation rate, the fertilization rate, the cleavage rate and the blastocyst rate. Observed the morphological characteristics of spindles and chromosomes of the oocytes. Result In tartrazine treated group, the percentage of activated oocytes, the rate of fertilization, cleavage and blastocyst were all lower than control group; and the oocytes displayed barrel spindle, the chromosomes became scattered. Conclusion Tartrazine reduces the developmental potential of mice oocytes.
