摘要
目的探讨活性维生素D3——1,25-二羟维生素D3[1,25-(OH)2D3]对单侧输尿管梗阻(UUO)大鼠Erbin表达的影响。方法将96只大鼠采用随机数字表法随机分为正常对照组、干预组、假手术组和UUO模型组,每组24只。干预组和UUO模型组大鼠在左肾下极处游离并结扎左输尿管建立UUO模型,假手术组开腹后仅游离左输尿管,干预组在建模起给予活性维生素D30.5μg、花生油1 m L灌胃,每天1次,共计14 d,于术后1、3、7、14 d分别处死各组大鼠6只,采用免疫组织化学法检测各组大鼠Erbin、转化生长因子β1(TGF-β1)的表达。结果 UUO模型组大鼠Erbin、TGF-β1表达均明显高于正常对照组、假手术组,差异均有统计学意义(P<0.05);干预组大鼠Erbin表达明显高于模型组,TGF-β1表达明显低于模型组,差异均有统计学意义(P<0.01)。结论 1,25-(OH)2D3可能增加UUO大鼠肾组织Erbin的表达,抑制TGF-β1的表达,减轻肾脏纤维化。
Objective To investigate the effect of activated vitamin D3-1,25-(OH)2D3 on the Erbin expression in uni lateral ureteral obstruction (UUO)rat. Methods Ninety-six SD rats were randomly divided into the normal control group ,intervention group, sham operation group and UUO model group aeeording to the random number table method,24 cases in each group. The UUO model in the intervention group and the UUO model group was established by freeing and ligating the left ureter at the inferior polar of the left kidney,the left ureter in the sham operation group was only dissoeiated after lapatotomy ,while the intervention group was given aetivated vitamin D3 0.5μg and peanut oill mL by gavage from the beginning of model construction ,once daily,for 14 d. Six rates in each group were killed on postoperative 1,3,7,14 d. The immunohistochemieal method was used to deteet the expression of TGF-β1 and Erbin in each group. Results The expressions of Erbin and TGF-β1 in the UUO model group were significantly higher than those in the normal control group and sham operation group, the differenees were statistically significant (P〈0.05) ;the expression of Erbin in the intervention group was significantly higher than that in the model group,while the TGF-β1 expression was significantly lower than that in the model group, the difference was statistically significant (P〈0.01). Conclusion 1,25-(OH)2D3 may increase the Erbin expression in renal tissue of UUO rat,inhibits the TGF-β1 expression and alleviates renal interstitial fibrosis.
