摘要
利用液相色谱对淡紫拟青霉产生的色素进行分析,确定其为单一物质(保留时间为8.845 min),从而建立色素相对含量的测定方法。在此基础上对色素的发酵条件进行优化,并对色素的抑菌性能进行了初步研究。依据响应面法分析得到最佳发酵条件:Na NO_3质量浓度为0.53 g/L,K2HPO4质量浓度为0.41 g/L,Tween-80体积分数为0.17%,在此条件下,色素的产量是优化前的1.95倍。抑菌实验结果表明,该色素对于细菌有广谱抑制作用,对于部分真菌如头孢霉等也有很强的抑制作用。
The pigment produced by Paecilomyces lilacinus TD16 is identified as a single substance with the retention time of 8. 845 min on high-pressure liquid chromatography( HPLC). A quantitative method is established for determining the relative content of this pigment. Accordingly,the fermentation conditions for pigment production are also optimized based on Box-Behnken design( BBD). Under the optimized conditions,0. 53 g/L of Na NO_3,0. 41 g/L of K2HPO4 and 0. 17%( v/v) of Tween-80,the pigment yield is 1. 95 times higher than that before optimization.Antimicrobial tests indicate that the pigment of TD16 can inhibite all the bacteria in this study,including part of the fungi such as Cephalosporium,which is of potential application in industries.
