摘要
冬虫夏草[Cordyceps sinensis(Ber K.)Sacc.]菌培养过程中,蝙蝠蛾拟青霉菌株SH-1的菌丝产生粉红色变化的现象,粉红菌株与SH-1菌株形态上存在很大差异,为查明产生的原因,在单因素试验基础上考察培养时间、培养温度、光照、p H和装液量对粉红色变化的影响,通过响应面法优化冬虫夏草真菌的培养环境,分析影响粉红色变化的关键因素。结果表明,产生粉红变化的最适培养条件为培养时间80 d、培养温度20.55℃、p H 7.51、装液量250 m L,在此培养基条件下,粉红菌株ZH-1产生概率为35.14%;得出菌株ZH-1的最大得率培养条件为培养时间80 d、培养温度为22.25℃、p H 6.89、装液量250 m L,在此培养基条件下,粉红菌株ZH-1得率为8.95%。
A portion of normal fungi Paecilomyces hepia!i strain SH-1 changed pink color gradually during cultured Cordyceps sinensis (BerK.) Sacc. There were significant differences in the morphology of pink and SH-1 strains. For seeking the reason of emerging pink variation and obtaining the maximum of yield ratio of the pink fungi, five main factors including culture time, culture temperature, pH value, loading volume and light conditions were selected to analyze key factor of influencing pink change by response surface methodology on the basis of experimental results of single factors. The results showed that the optimum culture conditions of the strains SH-1 change to ZH-1 were culture time 80 d,culture temperature 20.55 ℃,pH value 7.51 ,loading volume 250 mL. Under such conditions,the change probability of ZH-1 was 35.14%. The optimum culture conditions of ZH-1 were culture time 80 d,culture temperature 22.25 ℃ ,pH value 6.89,loading volume 250 mL. Under these conditions, the yield of ZH-1 was 8.95%.
