摘要
研究花魔芋(Amorphophallus albus)愈伤组织诱导的最佳激素组合培养条件。以魔芋叶柄为外植体,采用4因素3水平正交试验研究2,4-D、NAA、6-BA与KT 4种激素对魔芋愈伤组织诱导的影响。结果表明,2,4-D、NAA、6-BA与KT对魔芋叶柄外植体膨大与愈伤组织形成具有显著效应。利于外植体膨大的最优激素组合为MS+3.0 mg/L 2,4-D+3.0 mg/L NAA+0.3 mg/L 6-BA+0.1 mg/L KT;利于愈伤组织积累的最优激素组合为MS+1.0 mg/L 2,4-D+3.0 mg/L NAA+0.3 mg/L 6-BA+1.0 mg/L KT。试验优化的激素组合条件下魔芋外植体膨大与愈伤诱导有较佳的表现,可用于花魔芋的培养。
Screen the optical plant hormone combination for callus induction of Amorphophollus albus. Selecting petiole as materials, four hormone combination of 2,4-D,NAA,6-BA and KT were researched on explants expanding and callus inducing by orthogonal test. Results showed that 2,4-D,NAA and 6-BA had significant effects on expanding rate and the four hormones all exerted extremely obvious effects on callus induction rate. The optimal hormone combination for expanding was MS+3.0 mg,/L 2,4-D+3.0 mg/L NAA+0.3 mg,/L 6-BA+0.1 mg/L KT and combination for callus accumulation was MS+1.0 mg./L 2,4-D+3.0mg/L NAA+0.3 mg/L 6-BA+1.0 mg/L KT,respectively. The hormone combinations obtaining higher expanding rate and callus induction rate were suitable for tissue culture of Amorphophallus albus.
