露花微繁技术体系研究

摘要 以温室中生长的露花(Mesembryanthemum cordifolium L.f.)幼嫩茎段为外植体,利用正交试验等方法研究了露花微繁技术体系。结果表明,用70%~75%乙醇浸泡30 s、0.1%升汞浸泡6 min对外植体消毒灭菌就能达到理想的效果;对于无顶芽茎段,丛生芽增殖最适培养基是MS+NAA 0.05 mg/L+6-BA 0.9~1.5 mg/L+GA30.3 mg/L+蔗糖30 g/L+琼脂7 g/L;而对于有顶芽茎段要达到相同增殖预期,则在其他成分不变的前提下,需要6-BA浓度达到1.5 mg/L。因此,无顶芽茎段更适合作为丛生芽增殖的培养材料;生根最适培养基是1/2 MS+IBA 0.2 mg/L+蔗糖30 g/L+琼脂7 g/L。 Using the tender stem growing in greenhouse as explants, orthogonal test was applied to study the micropropagation technique system of Mesembryanthemum cordifolium L. f.. The results showed that dipping in 70% to 75% alcohol for 30 s and 0.1% mercuric chloride for 6 min would had idea sterilization effect. For tender stem without terminal bud as explants,the optimal medium for clump bud proliferation was MS+0.05 mg / L NAA+0.9 to 1.5 mg / L 6-BA+0.3 mg / L GA3+30 g / L sugar+7 g / L agar; while for tender stem with terminal bud, the 6-BA concentration should be 1.5 mg / L. Therefore, tender stem without terminal bud was more suitable explants for clump bud proliferation. The optimal medium for rooting was 1 / 2 MS+0.2mg / L IBA+30 g / L sugar+7 g / L agar.
