摘要
目的 探讨赖氨酸特异性去甲基化酶1 (LSD1)在胶质瘤及非肿瘤脑组织中的表达并分析其临床意义.方法 选取河北大学附属医院2014年1月至2015年7月收治的50例新鲜胶质瘤标本和10例非肿瘤脑组织标本.采用反转录-聚合酶链反应(RT-PCR)法检测LSD1 mRNA的表达水平,采用免疫组织化学法检测LSD1蛋白的表达水平.结果 LSD1 mRNA相对表达量在脑组织中为2.32 ×10^-3±2.23 ×10^-3,Ⅱ级胶质瘤为6.86x10^-3±2.72x10^-3,Ⅲ级胶质瘤为4.58×10^-2±6.82×10^-3,Ⅳ级胶质瘤为5.86×10^-2±1.31×10^-2;LSD1蛋白在脑组织中阳性标记指数为(7.50±4.33)%,Ⅱ级胶质瘤中为(16.21±4.82)%,Ⅲ级胶质瘤中为(40.24±5.46)%,Ⅳ级胶质瘤中为(48.26±6.91)%.结论 LSD1在胶质瘤中高表达,并且其表达量随胶质瘤级别的增高而增高。
Objective To investigate the expression and significance of lysine-specific demethylase1 (LSD1) in human gliomas.Methods Fifty glioma tissues and ten non-tumor brain tissues were collected from The Affiliated Hospital of Hebei University.Reverse transcriptase-polymerase chain reaction (RT-PCR) was applied to detect mRNA expression of LSD1.We detected the expression of LSD1 at protein level by using immunohistochemistry.Results The expression levels of LSD1 mRNA in non-tumor brain was 2.32 × 10^-3 ± 2.23 × 10^-3,in glioma grade 2 was 6.86 × 10^-3 ± 2.72 × 10^-3,in glioma grade 3 was 4.58 × 10^-2 ±6.82 × 10^-3,in glioma grade 4 was 5.86 × 10^-2 ± 1.31 × 10^-2;the positive marker index of LSD1 protein in brain was (7.50 ±4.33)%,in glioma grade 2 was (16.21 ±4.82)%,in glioma grade 3 was (40.24 ±5.46)%,in glioma grade 4 was (48.26 ±6.91)%.Conclusion LSD1 was highly expressed in gliomas.The expression level of LSD1 was related with pathological grades.
