乳腺癌中DNA聚合酶-β的表达及其对乳腺癌细胞迁移的影响 被引量:2

The expression of DNA polymerase-β in breast cancer and its effect on migration of breast cancer cells

摘要 目的 观察DNA聚合酶-β(DNA pot-β)在乳腺癌中的表达及其对乳腺癌细胞迁移能力的影响.方法 实时定量聚合酶链反应(Real-time PCR)检测75例乳腺癌组织及配对的癌旁组织中DNA pol-β的表达,分析其与淋巴结转移的关系.转染siRNA-DNA pol-β以观察DNA pol-β对乳腺癌细胞迁移能力的影响.结果 同癌旁组织比较,癌组织中DNA pol-β的表达显著升高(2.80 ±2.27比3.49±2.56,P<0.01),淋巴结转移与DNA pol-β的表达呈正相关(χ^2=5.580,P<0.05).同空白对照组(23.0±4.0)和阴性对照组(20.1±3.8)比较,siRNA-DNA pol-β转染组(10.1±2.5) MCF-7细胞的迁移能力显著下降(P<0.05).结论 DNA pol-β在乳腺癌中表达上调,可能是乳腺癌细胞转移的机制之一。 Objective To investigate the expression of DNA polymerase-β (DNA pol-β) in breast cancer and its effect on migration of breast cancer cells.Methods The mRNA level of DNA pol-β in breast cancer tissues and paired paracancerous tissues were detected by real-time quantitative polymerase chain reaction (Real-time PCR) and the relationship between DNA pol-β expression and lymph node metastasis in patients with breast cancer were analyzed.The effect of DNA pol-β on migration of breast cancer ceils was investigated.Results The expression of DNA pol-β was up-regulated in breast cancer compared with adjacent breast tissues (2.80 ± 2.27 vs.3.49 ± 2.56,P 〈 0.01).The increased expression of DNA pol-β in breast cancer tissues was positively associated with lymph node metastasis (χ^2 =5.580,P 〈0.05).MCF-7 cells transfected with siRNA-DNA pol-β (10.1 ±2.5) showed decreased migration compared with control group (23.0 ± 4.0) and negative siRNA group (20.1 ±.3.8) (P 〈 0.05).Conclusion The up-regulation of DNA pol-β in breast cancer tissue is a possible mechanism responsible for the metastasis of breast cancer cells.
