摘要
目的 探讨长链非编码RNA(lncRNA)肌动蛋白纤维相关蛋白1-反义RNA1(AFAP1-AS1)在胃癌中的表达及其临床意义,并在胃癌细胞株中验证其细胞学功能.方法 采用实时荧光定量聚合酶链反应(RT-qPCR)检测我院60例胃癌患者的癌及对应癌旁组织中AFAP1-AS1的水平,分析AFAP1-AS1的表达与临床病理特征的关系;进一步使用RT-qPCR检测5个不同的胃癌细胞株中AFAP1-AS1的表达水平.对于高表达AFAP1-AS1的细胞株使用慢病毒载体下调基础表达.进而检测下调AFAP1-AS1后对细胞增殖、凋亡和侵袭能力的影响.结果 胃癌组织中AFAP1-AS1的相对表达量△Ct水平显著高于癌旁组织水平(6.349±1.966比3.065±1.015,P<0.01),且其表达与组织学分级、TNM分期、淋巴结转移水平有关(P<0.05);胃癌细胞株MGC-803及SGC-7901高表达(2.239±0.020及2.488±0.021),有效干扰AFAP1-AS1的表达后可以显著抑制MGC-803及SGC-7901细胞的增殖、侵袭能力并促进凋亡(P<0.01).结论 AFAP1-AS1在胃癌组织中高表达.沉默AFAP1-AS1后可以显著抑制胃癌细胞的增殖、侵袭能力并促进凋亡。
Objective To explore the expression and clinical significance of long non-coding RNA (IncRNA) actin filament-associated protein 1-antisense RNA 1 (AFAP1-AS1) in gastric cancer (GC),and to verify its function in GC cells.Methods The levels of AFAP1-AS1 were detected in 60 couples of GC specimens,corresponding adjacent tissues and in 5 gastric cancer cell lines using the real-time quantitative polymerase chain reaction (RT-qPCR).The relationship between the AFAP1-AS1 and clinicopathological parameters was analyzed.Cells (MGC-803 and SGC-7901) with highly expressed AFAP1-AS1 were down-regulated lentiviral short hairpin RNA (shRNA) directed against AFAP1-AS1.The effects of the AFAP1-AS1 knockdown on cell proliferation,invasion and apoptosis were performed by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyhetrazolium bromide (MTT)method,flow cytometry with propidium iodide (PI)/annexin V double staining and Transwell.Results The ACt level of AFAP1-AS1 expressed in tumor tissue was significantly higher than that in paracarcinoma tissues (6.349 ± 1.966 vs.3.065 ± 1.015,P 〈0.01),and related with TNM staging,lymph node metastasis,and tumor differentiation (P 〈 0.05),respectively.AFAP1-AS1 was highly expressed in MGC-803 and SGC-7901 gastric cancer cell lines (2.239 ± 0.020,and 2.488 ± 0.021).MTT assay revealed the absorbance values of AFAP1-AS1 knockdown group were lower than in the control group at 48 and 72 h(P 〈0.01).Moreover,there was higher apoptosis rate but less transmembrance cell number.Conclusion The expression of AFAP1-AS1 was higher in both GC tissues and cells,respectively.Knockdown of AFAP1-AS1 may inhibit the cell proliferation and invasion but induce the apoptosis of GC cells.
