摘要
肥胖等代谢性疾病的发生发展与脂肪细胞的发育和功能异常有着非常密切的联系.现阶段,调控间充质干细胞及前脂肪细胞向成熟脂肪细胞分化的分子机制的研究已成为热点.许多新的证据表明,一些酶、细胞因子、蛋白超家族等作为共调节因子级联激活或抑制转录因子进而调控成脂基因的表达,在成脂分化中起关键性作用.早期学界对成脂分化的认识大多集中在前脂肪细胞终末分化阶段,而对于间充质干细胞向前脂肪细胞定向及前脂肪细胞的早期分化的转录调控机制所知甚少.基于近年来对调控间充质干细胞及前脂肪细胞成脂分化的分子及机制研究的逐步深入,本文就转录因子及共调节因子调控间充质干细胞及前脂肪细胞成脂分化的研究新进展作一综述。
The occurrence and development of metabolic diseases,such as obesity,have very close relationship with the development or abnormal function of fat cells.At this stage,the molecular mechanism of regulation of mesenchyeal stem cells and the differentiation of fat cells to mature adipocytes has become a hot spot.Many new evidence suggests that some co-regulatory factors such as enzymes,cytokines,protein superfamily stimulate signaling cascades that lead to activation or repression of transcription factors and regulate lipid gene expression,which plays a critical role in adipogenic differentiation.The early knowledge of adipogenic differentiation were mostly concentrated in the terminal differentiation stage of preadipocytes,but the mechanism of commitment of mesenchymal stem cell and the early stage of transcriptional regulation in preadipocytes was poorly understood.Based on the deep research in recent years on the molecular mechanism of regulating mesenchymal stem cells and preadipocytes into adipocytes,new progress on regulation of transcription factors and co-regulatory factors in the differentiation of fat are reviewed in this paper.
