盐酸多柔比星神经节介入对脑缺血影响的实验研究

Experimental study on the influence of doxorubicin hydrochloride on neurovascular intervention in cerebral ischemia reperfusion of rats

摘要 目的研究盐酸多柔比星神经节介入对脑缺血再灌注模型大鼠血管内皮生长因子(VEGF)、过氧化物歧化酶(SOD)及丙二醛(MDA)水平的影响。方法30只Wistar大鼠按体重随机分为模型组、实验组以及空白组,每组各10只。用Pulsinelli四血管阻断法,建立脑缺血再灌注模型。实验组于术前48 h,开始于椎间孔缓慢注射盐酸多柔比星1 mg·kg-1,模型组与空白组注射同等剂量生理盐水q12 h。以酶联免疫吸附法(ELISA)测定大鼠脑组织内VEGF、SOD及MDA水平。结果在给药12,24 h的神经功能评分,与空白组的0分比较,模型组分别为(2.41±0.25),(1.98±0.23)分,明显增加,2组比较差异有统计学意义(P<0.05);与模型组比较,实验组分别为(1.58±0.19),(1.31±0.15)分,明显降低,2组比较差异有统计学意义(P<0.05)。模型组在给药12,24 h,脑组织VEGF表达分别为124.63±15.63,120.57±12.84,血清中丙二醛水平分别为(3.56±0.38),(3.85±0.35)mmol·mg^(-1);实验组在给药12,24 h,脑组织VEGF表达分别为102.26±12.38,100.47±11.91,血清中丙二醛水平分别为(2.97±0.35),(3.25±0.35)mmol·mg^(-1),较模型组明显降低,2组比较差异有统计学意义(P<0.05)。在给药12,24 h,模型组血清中SOD分别为(120.48±15.03),(119.37±12.58)U·mg^(-1);实验组血清中SOD水平分别为(182.73±19.49),(181.57±18.63)U·mg^(-1),实验组较模型组明显升高,2组比较差异有统计学意义(P<0.05)。结论盐酸多柔比星神经节介入通过改善氧化应激反应以及炎症反应,从而保护脑缺血再灌注大鼠脑组织缺血再灌注损伤。 Objective To study the influence of doxorubicin hydrochlo- ride intervention on cerebral ischemia reperfusion in rats. Methods Thirty Wistar rats were randomly divided into the model group, experi- mental group and blank group, with 10 rats in each group. The cerebral ischemia reperfusion model was established by using Pulsinelli four vas- cular occlusion method. The rats of experimental group in the preopera- tive 48 h began using doxorubicin hydrochloride 1 mg·kg-1 for injection in the slow injection of the intervertebral foramen ; the rats of model group and the blank group was treated normal saline with the same method as the experimental group, q12 h. After the intervention, the levels of vas- cular endothelial growth factor (VEGF), superoxide dismutase (SOD) and the lipid peroxidative product -malondialdehyde (MDA) in the brain tissues of rats were determined by ELISA. Results Compared to blank group in nerve function score (0) at 12 and 24 h, the neural function score in model group with (2.41 ±O. 25 ), ( 1.98 ±0. 23) score were higher ( P 〈 0. 05 ). Compared to model group, the nerve function score in experimental group with ( 1.58 ±0. 19), ( 1.31 ±0. 15) at 12 h and 24 h were lower(P 〈0. 05). The expression of VEGF in model group were 124. 63 ± 15.63,120.57 ± 12. 84 ; and the level of MDA in model group were (3.56 ± 0. 38), (3.85 ± 0. 35 ) mmol·mg-1 at 12 h and 24 h. But the expression of VEGF in experimental group were 102.26 ± 12. 38, 100. 47 ±11.91 and level of MDA in experimental group were (2.97 ± 0.35 ), (3.25 ± 0.35 )mmol·mg-1 at 12 h and 24 h, compared with the model group, the indexes in the experimental group were lower, these difference was significantly compared two groups(P 〈0. 05). Level of SOD serum in model group were ( 120. 48 ± 15.03) and ( 119. 37 ± 12.58) U ·mg-1 and in experimental group were (182. 73 ±19.49) and (181.57 ±8.63) U·mg-1 at 12 h and24 h, com- pared with the model group, SOD in the experimental group was higher(P 〈 0. 05 ). Conclusion The protective effects of doxorubicin hydrochloride on the cerebral ischemia reperfusion injury in rats by improving the oxidative stress and in- flammatory response.
