摘要
目的探讨大蒜素(allicin)对多氯联苯(PCBs)诱导小鼠骨骼肌细胞氧化应激及葡萄糖转运蛋白-4(GLUT-4)表达的影响。方法 50只健康成年小鼠随机分为对照、多氯联苯、大蒜素(30、40、60 mg/kg)组,每组10只;连续给予30 d,取小鼠骨骼肌(股薄肌和肱三头肌),显微观察骨骼肌细胞形态,化学比色法检测骨骼肌组织丙二醛(MDA)含量及总超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶(CAT)活力,免疫组化检测GLUT-4表达量。结果多氯联苯组小鼠骨骼肌纤维排列紊乱,肌细胞肿胀,横纹基本消失;与对照组比较,多氯联苯组小鼠股薄肌和肱三头肌组织中MDA含量[分别为(19.15±2.73)与(22.19±3.72)nmol/mgprot]明显增加(P<0.05),股薄肌与肱三头肌T-SOD、GSH-Px、CAT酶活力[分别为(110.11±15.55)、(48.55±7.49)、(5.01±1.79)与(102.32±16.80)、(45.42±10.73)、(5.43±1.12)U/mgprot]明显下降(P<0.05);与多氯联苯组比较,40 mg/kg大蒜素组小鼠股薄肌与肱三头肌组织中MDA含量[分别为(12.17±2.18)与(13.00±2.18)nmol/mgprot]明显下降,T-SOD、GSH-Px、CAT酶活力[分别为(145.80±18.41)、(59.16±11.18)、(7.27±2.16)与(158.58±5.02)、(75.40±5.03)、(8.35±0.86)U/mgprot]明显升高(P<0.05);与对照组比较,多氯联苯组小鼠骨骼肌GLUT-4表达减少;与多氯联苯组比较,大蒜素组小鼠骨骼肌GLUT-4表达明显升高。结论大蒜素对多氯联苯诱导骨骼肌细胞氧化应激及GLUT4蛋白表达具有一定的拮抗作用。
Objective To explore the effect of allicin on oxidative stress and glucose transporter 4 (GLUT-4) expression induced by polychlorinated biphenyls (PCBs) in skeletal muscle tissue of mice. Methods Fifty mice were randomly divided into a control group, a PCBs exposure group, and three PCBs + allicin exposure groups ( 30,40, and 60 mg/kg). At the end of 30 days' treatment, the morphology of skeletal muscle tissue was obsereved; the content of maleic dialdehyde (MDA) and the activities of total superoxide dismutase (T-SOD),catalase (CAT), and glutathione peroxidase (GSH-Px) in skeletal muscle tissue were detected;and the expression of GLUT-4 was also measured with immunohistochemistry. Results Compared with the control group, the disordered arrangement, cell swelling, and disappearance of cell stripes were observed in skeletal muscle tissue of PCBs exposure group ; the contents of MDA increased obviously ( 19. 15 ± 2. 73 and 22. 19 ±3.72 nmol/mg prot in gracilis and triceps, both P 〈 0. 05 ) ; and the activities of the T-SOD ,GSH-Px,and CAT decreased significantly ( 110. 11 ± 15.55,48.55 ±7.49 ,and 5.01 ± 1.79 U/mg prot in gracilis, 102. 32± 16. 80,45. 42 ± 10. 73, and 5.43 ± 1.12 U/rag prot in triceps ; all P 〈 0. 05 ). Compared with the PCBs exposure group, the contents of MDA obviously decreased in skeletal muscle tissue of the mice with PCBs + 40 mg/kg allicin exposure ( 12. 17 ± 2. 18 and 13.00 ± 2. 18 nmol/mg prot in gracilis and triceps, both P 〈 0. 05 ) and the activities of the T-SOD, GSH-Px, and CAT significantly increased ( 145.80 ± 18.41,59. 16 ±11.18, and 7.27 ± 2. 16 U/mg prot in gracilis,158.58 ± 5.02,75.40 ± 5.03, ans 8.35 ± 0. 86 U/mg prot in triceps; all P 〈 0. 05 ). Compared with the control group, the expression of GLUT-4 decreased in the PCBs exposure group, but obviously increased in the PCBs + allicin exposure group. Conclusion Allicin has antagonistic effect on the expression of GLUT-4 in cytoplasm of skeletal muscle cells through its anti-oxidation in mice.
