摘要
目的探讨溶血和乳糜微粒对ALT检测的影响。方法根据中华人民共和国卫生行业标准(WS/T)和美国临床和实验室标准研究院(CLSI)相关文件,进行干扰实验验证。结果干扰实验结果显示,样本在反复冻融1次以上和震荡2 min以上时,以及外源性干扰物血红蛋白(HB)>2.5 g/L时ALT检测均产生正干扰,乳糜微粒>5 g/L对ALT检测有明显负干扰。结论血红蛋白产生的是正干扰,乳糜微粒产生的是负干扰,通过评估溶血和脂血标本的干扰方向和干扰偏差的大小,对如何处理生物参考区间上限附近的标本具有指导作用。
Objective To investigate the effect of hemolysis and chylomicron on ALT detection. Methods Interference tests were conducted on the relevant documents from the health industry standards( WS / T) in the People’s Republic of China and the Clinical and Laboratory Standards Institute( CLSI). Results Interference tests showed that when samples were frozen and thawed for at least 1 time and shocked for more than 2 minutes,and when exogenous HB 〉 2. 5 g / L,positive interference was imparted on ALT detection. However,when chylomicron 〉 5 g / L,a significant negative interference was produced. Conclusion Hemoglobin makes positive interference and chylomicrons make negative interference. By evaluating the direction of interference and the size of the interference bias of the hemolysis and lipid blood sample,guidance is provided on dealing with the sample near the upper limit of the biological reference interval.
