摘要
目的:测定海南槟榔乙醇提取物中多酚和槟榔碱的含量。方法:多酚的含量分析采用分光光度法,以没食子酸为对照品,以FeCl3-K3[Fe(CN)6]为显色剂,在720nm处进行测定;槟榔碱的含量分析采用高效液相色谱法,色谱柱为C18柱,流动相组成为乙二胺、甲醇和乙腈,检测波长为215nm,流速1.0mL/min。结果:海南槟榔乙醇提取物中多酚的含量为10.54mg/g,线性范围为0.096~0.576μg/mL,平均回收率为101.40%,RSD为0.88%;海南槟榔乙醇提取物中槟榔碱的含量为1.935μg/g,RSD为1.8%。结论:该方法简单快捷,重现性好,适合于海南槟榔乙醇提取物中多酚和槟榔碱的含量测定。
Objective.. To establish a method for determination of total polyphenols and arecoline in ethanol extracts from Areca catechu. Methods.. The content of total polyphenols was determined by spectrophotometry at 215 nm, with the mixture solution of FeC18 and K3 [Fe(CN)6] as the color developing agent. The content of arecoline was analyzed by HPLC. Hypersil BDS C18 was used and the mobile phase was consisted of diethylamine, metanol and acetonitrile. The detection wave- length was 215 nm,and the flow rate was 1.0 mL/min. Results: The content of total polyphenols in ethanol extracts from Areca catechu was 10.54 mg/g. Calibration graphs were constructed in the range of 0.096-0.576 μg/mL. The average recoveries were 101.40%. The RSD of the methods was 0.88%. The content of arecoline was 1.935 μg/g, with the RSD of 1.8%. Conclusions: The method was brief, fast, accurate, and suitable for the content determination of total polyphenols and arecoline in ethanol extracts from Areca catechu.
