胸苷酸合成酶基因多态性与培美曲塞治疗海南黎族肺腺癌患者效果的相关性 被引量:2

摘要 目的探讨胸苷酸合成酶(TS)rs34743033(5'端非编码区28 bp重复序列)及rs34489327(3'非编码区的1 494 bp处6 bp核苷酸片段的缺失或插入)与培美曲塞治疗海南黎族肺腺癌患者疗效与预后的相关性。方法手术后经病理诊断确认海南晚期肺腺癌患者104例,采用PCR的方法检测5'端28 bp重复序列多态性;采用焦磷酸测序的方法检测3'端6 bp核苷酸片段的缺失或插入,随访记录患者的无进展生存时间和总生存期。结果 TS基因5'端非编码区rs34743033多态性位点3R/3R、2R/3R、2R/2R基因型在肺腺癌患者所占的比例分别为67.3%(70/104)、17.3%(18/104)、15.4%(16/104)。3'端非编码区rs34489327多态性位点+6 bp/+6 bp、+6 bp/-6 bp、-6 bp/-6 bp基因型在肺腺癌患者中所占比例分别为53.8%(56/104)、34.6%(36/104)、11.5%(12/104)。2R/2R和2R/3R基因型患者分别为12.00和18.50个月,3R/3R基因型患者中位无复发生存时间和总生存时间分别为11.63和18.17个月,二者之间无显著相关;-6 bp/-6 bp和-6 bp/+6 bp基因型患者分别为12.00和18.70个月,+6 bp/+6 bp基因型患者中位无复发生存时间和总生存时间分别为11.50和19.17个月,二者之间无显著相关。结论 TS基因rs34743033及rs34489327多态性可能与培美曲赛治疗海南黎族肺腺癌的疗效和预后无关。
