摘要
目的探讨色素上皮衍生因子(PEDF)对脑胶质瘤U87MG细胞增殖与凋亡的影响。方法分组培养U87MG细胞,对照组不添加PEDF蛋白,干预组分别添加80nmol/L、160nmol/L、320nmol/L的PEDF蛋白,采用MMT法检测不同时间点各组细胞增殖抑制率,采用流式细胞仪检测各组细胞凋亡率并进行比较。结果同一时间点,各浓度PEDF干预组的细胞增殖抑制率均明显高于对照组,差异有统计学意义(P<0.05);各浓度PEDF干预组间比较差异均有统计学意义(P<0.05);相同浓度PEDF环境下,各时间点间细胞增殖抑制率差异亦有统计学意义(P<0.05)。随着PEDF浓度增高,U87MG细胞凋亡率呈增高趋势。结论 PEDF能够抑制脑胶质瘤U87MG细胞增殖并能促其凋亡,其抑制增殖水平与浓度、作用时间相关,促进凋亡水平与浓度相关。
Objective To investigate the effects of pigment epithelium-derived factor (PEDF) on cell proliferation and apoptosis in U87MG cells of brain glioma. Methods U87MG ceils were primarily cultured. The cells without PEDF were used as control group. The cells with 80nmol/L, 160nmol/L and 320nmol/L PEDF were used as intervention groups. The inhibitory rates of poliferation of U87MG cells were observed by MMT assay. The apoptotic rates were detected by flow cytometry. Results The inhibitory rates of proliferation of Ug7MG cells in PEDF intervention groups were higher than that in control group at a certain time(P〈0.05). There were statistics differences among the PEDF intervention groups (P〈0.05). There were statistics differences among the PEDF intervention groups at the certain concentration of PEDF in different times(P〈0.05). The apoptosis rates of U87MG cells induced by PEDF were increased by de grees with augmentation of PEDF concentration. Conclusion PEDF may inhibit proliferation and induce apoptosis in brain glioma U87MG cells in vitro. The inhibition of proliferation may be associated with PEDF concentration and intervention time. The induction of apoptosis may be dependent on certain concentration of PEDF.
