摘要
目的:研究蒙药忠伦阿汤对胶原诱导性关节炎(CIA)模型大鼠免疫调节机制的影响。方法:本研究首先利用牛Ⅱ型胶原建立SD大鼠CIA模型,造模成功后随机分为正常组,模型组,蒙药高、中、低剂量组及雷公藤组。测量不同时间点大鼠体质量、关节炎指数,测量治疗后各组大鼠胸腺指数、脾脏指数、小肠派氏小结计数;对各组大鼠踝关节组织进行HE染色,光镜下观察踝关节组织病理学改变;ELISA检测各组大鼠血清干扰素-γ(IFN-γ)、转化生长因子-β(TGF-β)细胞因子及抗CⅡ抗体的含量。结果:各治疗组CIA大鼠体质量、关节炎指数、胸腺指数、脾脏指数、小肠派氏小结计数、大鼠踝关节组织病理学改变以及血清中IFN-γ、TGF-β、抗CⅡ抗体的含量均得到改善,与模型组比较,雷公藤组和蒙药忠伦阿汤高剂量组改善最为明显(P<0.05,P<0.01)。结论:蒙药忠伦阿汤对CIA大鼠具有显著的免疫调节作用,其作用机制与阳性对照药雷公藤多苷相当。
Objective:To study the influence of Mongolian medicine Zhongluna Decoction on immune regulation mechanism of collagen-induced arthritis(CIA) rat models.Methods:CIA rat models were established by using bovine collagen type I,and then divided into control group,CIA model group and high,middle low dosage of Mongolian medicine groups,and tripterygium glycosides group.The body weight,arthritis index of rats in each group at different time-points were determined.After treatment,thymus index,spleen index,and counts of Peyer's patches of rats in each group were measured.Pathological changes in ankle joint tissue of rats in each group were observed by using HE staining under light microscope.The contents of IFN-γ,TGF-β,and specific-c I antibody in serum of rats in each group were tested by using ELISA.Results:The body weight,the arthritis index,thymus index,spleen index,and counts of Peyer's patches,pathological changes,and contents of IFN-γ,TGF-β,and specific-c I antibody in serum of rats in treatment groups were all improved.Compared with model group,the improvements in high dosage of Mongolian medicine group and tripterygium glycosides group were more significant(P〈0.05,P〈0.01).Conclusion:Mongolian medicine Zhongluna Decoctionhas a significant immunomodulatory effect on CIA rats,and the mechanism is the same as tripterygium glycosides.
