朝医鹿茸大补汤对哮喘小鼠气道炎性反应的影响

Effects of traditional Korean medicine Lurong Dabu Decoction on airway inflammation in mice with asthma

目的:探讨朝医鹿茸大补汤对哮喘小鼠气道炎性反应的影响。方法:BABL/c小鼠40只随机分成5组:正常对照组,哮喘模型组,鹿茸大补汤低、高剂量组,地塞米松治疗组,每组8只。采用ELISA法检测BALF中炎性细胞分类、计数及炎性细胞因子白细胞介素-4(IL-4)、白细胞介素-5(IL-5)、白细胞介素-13(IL-13)、干扰素-γ(IFN-γ)含量;Western blot检测肺组织中磷酸化p38MAPK及NF-κB p65蛋白的变化;左肺组织行苏木精-伊红(HE)染色及NF-κB p65免疫组织化学染色。结果:与正常对照组比较,哮喘模型组小鼠支气管BALF中炎性细胞计数增多,IL-4、IL-5、IL-13水平增高(P<0.01),IFN-γ水平降低(P<0.01);肺组织磷酸化p38MAPK、NF-κB p65蛋白表达水平升高(P<0.01)。与哮喘模型组比较,朝医鹿茸大补汤低、高剂量组和地塞米松治疗组小鼠BALF中炎性细胞计数、IL-4、IL-5、IL-13水平和肺组织磷酸化p38MAPK及NF-κB p65蛋白表达水平均明显低,IFN-γ水平明显上升(P<0.05,P<0.01)。结论:朝医鹿茸大补汤对小鼠哮喘具有保护作用,其机制可能部分与抑制磷酸化的p38 MAPK以及NF-κB信号通路有关。

Objective：To investigate the effects of Lurong Dabu Decoction of traditional Korean medicine on airway inflammation in asthmatic mice.Methods：Forty BALB/c mice were divided into five groups as control group,asthma group,Lurong Dabu Decoction low,high dose group and dexamethasone group.The numbers of inflammatory cells and the concentrations of IL-4,IL-5,IL-13,IFN-γ in BALF were measured by ELISA,p38 MAPK and NF-κB p65 protein levels from the right lung tissues were detected by Western blot.Pathological examination from the left lung tissue was investigated by hematoxylin and eosin stain,and the NF-κB p65 phosphorylation in lung tissue was detected by immunohistochemical staining.Results：The asthmatic mice developed the following pathophysiological features：the numbers of inflammatory cells were increased,the levels of IL-4,IL-5,IL-13 in BALF and the lung tissues were increased（P〈0.01）,and the levels of IFN-γ were reduced（P〈0.01）,the expression of phosphorylation p38 MAPK and NF-κB p65 protein in lung tissues were also increased（P〈0.01）.Lurong Dabu Decoction low and high groups,dexamethasone group significantly reduced inflammatory cells and IL-4,IL-5,IL-13 levels in BALF,decreased p38 MAPK phosphorylation in lung tissue and reduced airway inflammatory cell recruitment into airway,increased IFN-γ levels in BALF（P〈0.05,P〈0.01）.Conclusion：Lurong Dabu Decoction of traditional Korean medicine attenuates inflammation response of asthma through suppression of p38 MAPK and NF-κB signaling pathway activation.
