补肾养血法对超促排卵大鼠子宫自然杀伤细胞亚型与分类的研究 被引量:4

摘要 目的:探讨补肾养血法对超促排卵后孕鼠子宫内膜CD56+NK蛋白表达及NK1/NK2/NK3/NKr1细胞免疫应答平衡影响。方法:建立超促排卵后孕鼠模型,以补肾养血方干预孕鼠,提取子宫内膜中子宫自然杀伤(u NK)细胞上清液,应用ELISA法检测上清液中CD56+NK细胞、干扰素-γ(INF-γ)、肿瘤坏死因子-α(TNF-α)、白细胞介素-4(IL-4)、白细胞介素-5(IL-5)、转化生长因子-β(TGF-β)、白细胞介素-10(IL-10)蛋白表达改变。结果:超促排卵可降低CD56+NK细胞及TGF-β、IL-10表达(P<0.05),提高INF-γ、TNF-α、IL-4、IL-5的表达(P<0.05),使蛋白表达从NK3、NKr1免疫营养向NK1、NK2细胞免疫损伤偏移;中药补肾养血方可显著提高CD56+NK细胞、TGF-β、IL-10蛋白表达(P<0.05),显著降低INF-γ、TNF-α、IL-4、IL-5蛋白表达(P<0.05),促进NK1、NK2免疫损伤向NK3、NKr1免疫营养方向偏移。结论:补肾养血法可以通过增加CD56+NK细胞表达,调节子宫内膜NK1/NK2/NK3/NKr1细胞免疫应答平衡,提高子宫内膜容受性从而保证早期的妊娠稳定。 Objective:To study effects of ‘invigorating kidney and nourishing blood method' on expression of CD56^+ NK protein and NK1/NK2/NK3/NKr1 cell immune response balance in inl uent super stimulate ovulation rats.Methods:The rats were subjected to establish the super stimulate ovulation model.The fertilized rats were treated with ‘invigorating kidney and nourishing blood method'.Then,the levels of CD56^+,INF-γ,TNF-β,IL-4,IL-5,TGF-α and IL-10 in u NK cells supernatant of endometrium were detected with ELISA assay.Results:Super stimulate ovulation decreased the expression of CD56^+,TGF-β,IL-10 and increased the expression of INF-γ,TNF-β,IL-4,IL-5(P〈0.05),with the result that the protein expression changes from immune nutrition function cells like NK3,NKr1 to immune injury function cells like NK1 and NK2.The ‘invigorating kidney and nourishing blood method' increased the expression of CD56^+,TGF-β,IL-10 and decreased the expression of INF-γ,TNF-α,IL-4,IL-5(P〈0.05),with the result that the protein expression changes from immune injury function cells like NK1 and NK2 to immune nutrition function cells like NK3 and NKr1.Conclusion:The ‘invigorating kidney and nourishing blood method' could enhance CD56^+ NK cells expression and regulate NK1/NK2/NK3/NKr1 cells immune response balance in endometrium,improving endometrial reception that helps to maintain stability of pregnancy.
