刺山柑总生物碱对系统性硬皮病小鼠核心蛋白多糖等表达的影响 被引量:2

Effects of Capparis spinosa L.total alkaloids on expression of decorin,TNF-α,and PDGF-AB in mice with systemic scleroderma

摘要 目的:观察刺山柑总生物碱对系统性硬皮病(SSc)小鼠核心蛋白多糖(Decorin)、肿瘤坏死因子-α(TNF-α)及血小板源生长因子-AB(PDGF-AB)表达的影响。方法:BALB/c小鼠背部注射盐酸博莱霉素建立SSc模型,给予刺山柑总生物碱低、中、高剂量(225、450、900mg/kg)进行干预后,ELISA法检测小鼠皮肤Decorin、TNF-α及血清PDGF-AB表达水平。结果:刺山柑总生物碱中、高剂量可升高小鼠皮肤Decorin水平,降低TNF-α水平(P<0.05,P<0.01);对血清PDGF-AB含量无显著影响。结论:刺山柑总生物碱对SSc小鼠Decorin和TNF-α的异常表达具有一定调节作用。 Objective:To investigate the effects of Capparis spinosa L.total alkaloids on the expression of decorin,tumor necrosis factor-α(TNF-α) and platelet derived growth factor-AB(PDGF-AB) in mice with systemic scleroderma(SSc).Methods:Mice models with SSc were established by repeated local injections of bleomycin in the back of BALB/c mice.After giving Capparis spinosa L.total alkaloids at low dosage(225mg/kg),middle dosage(450mg/kg) and high dosage(900mg/kg),the expression level of decorin,TNF-α and PDGF-AB in serum in the skin of mice were detected by ELISA.Results:The level of decorin was increased and TNF-α was decreased(P〈0.05,P〈0.01) after administration of middle dosage(450mg/kg) or high dosage(900mg/kg) of Capparis spinosa L.total alkaloids;the level of PDGF-AB wasn't af ected.Conclusion:Capparis spinosa L.total alkaloids are effective in regulating abnormal expression of decorin and TNF-α in SSc.
