白英抑瘤作用有效部位的初步筛选 被引量:16

Preliminary screening on effective part of tumor inhibitory effect from solanum lyratum

摘要 目的:从白英不同组分[白英总碱(SLTA)、白英总皂苷(SLTG)、SLTA+SLTG、白英总提物]中筛选出抑瘤作用的主要有效部位,并通过对人癌细胞体外增殖的抑制作用进行验证。方法:首先建立S-180肉瘤和艾氏腹水瘤(EAC)荷瘤小鼠模型,给予阳性对照药及白英不同组分进行干预,分别以抑瘤率及小鼠的生命延长率为主要检测指标,比较组间抗肿瘤药效强度,选出白英抗肿瘤有效部位。选取人肝癌细胞SMMC-7721、人肺腺癌细胞A549、人胃癌细胞BGC-803、人宫颈癌细胞He La、人乳腺癌细胞MDA-MB-231多种人癌细胞作为研究对象,以紫杉醇为阳性对照药物,通过MTT法检测药物对癌细胞的抑制率,并计算相应的半数抑制浓度(IC50)值。同时显微镜下观察在给予IC50浓度的样品处理时细胞数目及形态的改变。结果:白英不同组分均有一定的抗肿瘤作用,其中SLTA的抑瘤作用最强,以中剂量(8g/kg)效果最好,对S-180荷瘤小鼠的抑瘤率为54.05%,对EAC荷瘤小鼠的生命延长率为31.71%。SLTA样品对多种人癌细胞均有一定的抑制作用,对人肝癌细胞SMMC-7721、人肺腺癌细胞A549、人胃癌细胞BGC-803、人宫颈癌细胞He La、人乳腺癌细胞MDA-MB-231的IC50分别为292.0、93.0、181.4、396.3、244.7mg/L。与空白对照组比较,半抑制下的细胞数目急剧下降,且均呈现萎缩的趋势。结论:SLTA为白英抑瘤作用的主要有效部位。 Objective:To screen the main effective part of antitumor from different components of solanum lyratum such as total alkaloids,total saponins,total alkaloids+total saponins,total extract,and verified by inhibitory effect on proliferation of human cancer cells in vitro.Methods:Sarcoma S-180 and ehrlich ascites carcinoma(EAC) tumor-bearing mouse models were established,and then models were intervened by positive control drugs and different components of solanum lyratum respectively.The between-group antitumor intensity was compared,and the effective parts of antitumor from solanum lyratum were selected out with tumor inhibitory rate and life extension rate of mice as the main indexes.The inhibition and half-inhibition concentration(IC50) of drugs on human hepatoma cell SMMC-7721,human lung adenocarcinoma cell A549,human gastric cancer cell BGC-803,human cervical cancer cell He La,and human breast cancer cell MDA-MB-231 were tested by using MTT method with paclitaxel as the positive control drug.The changes in cell number and form under the inhibiting of IC50 were observed by using microscope.Results:All the different components of solanum lyratum have certain anti-tumor effect,and the antitumor effect of total alkaloids is the strongest.Under the action of total alkaloids with strongest concentration,the anti-tumor rate on S-180 tumor-bearing mice is 54.05%,and life extension rate of EAC tumor-bearing mice is 31.71%.Total alkaloids have certain inhibition on various human tumor cells,and the IC50 value on human hepatoma cell SMMC-7721,human lung adenocarcinoma cell A549,human gastric cancer cell BGC-803,human cervical cancer cell He La,human breast cancer MDA-MB-231 cell were 292.0,93.0,181.4,396.3,244.7mg/L,respectively.Compared with the blank control group,cell number under inhibiting of IC50 decreased dramatically,and the cell morphology was shrunk.Conclusion:Total alkaloids of solanum lyratum is the main effective part for inhibiting tumor.
