白藜芦醇对LPS诱导HUVEC细胞线粒体跨膜电位及IL-6、TNF—α表达影响研究

摘要 目的探讨白藜芦醇对LPS诱导HUVEC线粒体跨膜电位及IL-6、TNF—α表达影响。方法将白藜芦醇分为160,80,40,20μg.L-1四个浓度组,同时给予LPSI?g/ml刺激24小时,CCK-8法检测细胞活力,荧光显微镜观察细胞线粒体跨膜电位,ELISA法检测IL-6、TNF-α含量。结果与模型组比较,白藜芦醇提高HUVEC,提高细胞线粒体跨膜电位,抑制IL-6、TNF—α表达,有统计学差异(P〈0.05)。结论白藜芦醇提高LPS刺激后HUVEC细胞存活率,可能与抑制炎症因子IL-6、TNF—α表达和提高细胞线粒体跨膜电位有关。 Objective To investigate the effects of resveratrol on the mitochondrial membrane potential and the expression of IL-6,TNF-α of the HUVEC cells induced by LPS.Metheds: HUVEC cells were stimulated by LPS 1?g/ml and 160,80,40,20μg.L21 resveratrol, after 24 hours ,cell viability was observed by CCK-8; mitochondrial membrane potential were tested by the fluorescence microscope',IL-6,TNF-α were detected by ELISA.Results; Compared with model group, resveratrol could increase the cell viability and the mitochondrial membrane potential,inhibited the IL-6,TNF-α expression ,with statistically Significant (P〈 0.05). Conclusion: Resveratrol could increase the viability of HUVEC cell induced by LPS, and the mechanism may be associated with inhibitig the IL-6, TNF-α expression and increasing the mitochondrial membrane potential.
